Plasma and joint Fluid Glypican-3 Are Inversely Correlated with the Severity Knee Osteoarthritis

Wanvisa Udomsinprasert et al.

Summary: This study found that the levels of glypican-3 in plasma and synovial fluid were negatively associated with the radiographic severity of knee osteoarthritis. Glypican-3 might serve as a potential biomarker for reflecting the severity of knee OA and could play a significant role in the pathophysiology of degenerative joint disease.

Genetic pathways disrupted by ENPP1 deficiency provide insight into mechanisms of osteoporosis, osteomalacia, and paradoxical mineralization

Nathan D. Maulding et al.

Summary: ENPP1 deficiency can lead to various bone diseases, including osteoporosis, rickets, and spinal ossification. Research has shown that ENPP1 deficient mice exhibit reduced Wnt ligand gene transcription in whole bone, increased transcription of soluble Wnt inhibitors in the liver and kidney, suggesting multiorgan inhibition of Wnt activity.

Vascular pathologies in chronic kidney disease: pathophysiological mechanisms and novel therapeutic approaches

Philip Duesing et al.

Summary: CKD and CVD are interlinked, with CKD increasing the risk of cardiovascular disease mainly due to accumulation of uremic toxins, chronic inflammation, and oxidative stress. Vascular alterations, especially atherosclerosis and vascular calcification, play a significant role in the association between CKD and cardiovascular mortality.

Sequence features, structure, ligand interaction, and diseases in small leucine rich repeat proteoglycans

Norio Matsushima et al.

Summary: Small leucine rich repeat proteoglycans (SLRPs) play crucial roles in the extracellular matrix by binding to collagens and interacting with cytokines, affecting various biological functions in cells. Mutations in SLRP genes are linked to human diseases, and investigating the effects of these mutations on structures can provide insights into disease mechanisms.

Growth factor delivery using extracellular matrix-mimicking substrates for musculoskeletal tissue engineering and repair

Robert C. H. Gresham et al.

Summary: Therapeutic approaches for musculoskeletal tissue regeneration commonly use growth factors to influence nearby cells for migration, proliferation, or differentiation. However, challenges in clinical translation include poor control and high dosages leading to side effects. The extracellular matrix plays a key role in regulating growth factor presentation and retention, impacting tissue regeneration.

Osteocytes but not osteoblasts directly build mineralized bone structures

Ke Wang et al.

Summary: This study found that osteocytes, not osteoblasts, directly build mineralized bone structures, and that osteoblasts in Dmp1-null or Hyp mice produce aberrant and poorly mineralized bone. Additionally, the constitutive expression of beta-catenin in osteocytes can recapitulate a similar osteomalacia phenotype as seen in Dmp1 null or Hyp mice. Thus, the conclusion is that osteocytes directly build bone, challenging the existing dogma.
