NK cells and ILCs in tumor immunotherapy

Simona Sivori et al.

Summary: Innate immune cells, especially NK cells, play an important role in tumor immunotherapy by controlling tumor growth and metastasis. Various approaches have been developed, such as using IL-15 to improve NK cell function and utilizing NK cell engagers to activate NK cells. Additionally, NK cells may express inhibitory checkpoints like PD-1, which can be targeted for therapeutic use against certain tumors.

P2X7 receptor and the NLRP3 inflammasome: Partners in crime

Pablo Pelegrin

Summary: ATP plays an essential role in cellular energetics and signaling. Extracellular ATP in inflammatory environments activates immune cells through P2X7 receptor, leading to the formation of inflammasomes and inflammatory cell death. Research on novel anti-inflammatory therapies targeting P2X7 receptor and NLRP3 pathway is ongoing.

Summary: NK cells play a crucial role in immune responses to tumors by killing target cells and producing cytokines, but they can become dysfunctional in the immunosuppressive tumor microenvironment. This article provides an overview of NK tumor infiltration and surveillance, as well as the mechanisms by which NK cells become dysfunctional.

Targeting NK Cells to Enhance Melanoma Response to Immunotherapies

Hansol Lee et al.

Summary: NK cells play a crucial role in the innate immune system and can enhance adaptive immune responses. The activation of NK cells is regulated by a balance of inhibitory and activating receptors. These cells are potent producers of proinflammatory cytokines and can elicit strong antitumor responses through secretion of perforin and granzyme B. Strategies to enhance NK cell responses are being evaluated in clinical trials to improve immunotherapy effectiveness.
