Design, synthesis, biological evaluation, and molecular docking study of thioxo-2,3-dihydroquinazolinone derivative as tyrosinase inhibitors

Nima Sepehri et al.

Summary: Tyrosinase inhibitors were designed and synthesized in this study, and a compound called 4m showed the best inhibitory activity. Kinetic evaluation revealed that 4m exhibited a mixed inhibition pattern against tyrosinase, and antioxidant evaluations showed moderate to weak potency.

Design, synthesis, in vitro and in silico studies of novel Schiff base derivatives of 2-hydroxy-4-methoxybenzamide as tyrosinase inhibitors

Aida Iraji et al.

Summary: The novel 2-hydroxy-4-methoxybenzohydrazide showed promising tyrosinase inhibitory activity, with 4d being the most potent inhibitor. In vitro experiments and molecular modeling analysis suggested that 4d could be a potential candidate for developing anti-tyrosinase agents.

The natural-based optimization of kojic acid conjugated to different thio-quinazolinones as potential anti-melanogenesis agents with tyrosinase inhibitory activity

Nima Sepehri et al.

Summary: Melanin and melanogenesis have dual roles in radioprotection and undesirable effects, while designed compounds show potent inhibition of melanogenesis and tyrosinase activity, along with mild antioxidant capacity. Compound 5j and 5h exhibit the best inhibitory activity and reduce melanin content in B16F10 cells with limited toxicity against malignant cells. The proposed binding mode of new inhibitors through molecular docking is consistent with structure-activity relationship analysis.

Design, synthesis, and biological evaluation of symmetrical azine derivatives as novel tyrosinase inhibitors

Somaye Karimian et al.

Summary: A series of symmetrical azine derivatives were designed, synthesized, and evaluated for their inhibitory activity against tyrosinase. Compound 3f showed effective tyrosinase inhibition, with a three-fold improvement in potency compared to kojic acid as the positive control. Kinetic study of compound 3f confirmed uncompetitive inhibitory activity towards tyrosinase.
