Site-Specific Chemoenzymatic Conjugation of High-Affinity M6P Glycan Ligands to Antibodies for Targeted Protein Degradation

Xiao Zhang et al.

Summary: A chemoenzymatic method enables efficient and site-specific conjugation of high-affinity M6P glycan ligands to antibodies, resulting in structurally well-defined antibody-M6P glycan conjugates. This method has potential applications for targeted degradation of membrane-associated proteins.

One-Pot Conversion of Free Sialoglycans to Functionalized Glycan Oxazolines and Efficient Synthesis of Homogeneous Antibody-Drug Conjugates through Site-Specific Chemoenzymatic Glycan Remodeling

Chong Ou et al.

Summary: ADCs utilize the specificity of antibodies to deliver toxic drugs for targeted cell killing, and site-specific conjugation methods are crucial for constructing homogeneous ADCs; a facile synthesis of functionalized glycan oxazolines from free sialoglycans and efficient endoglycosidase mutant for site-specific glycan transfer were reported for constructing homogeneous ADCs.

Diversity-Oriented Chemoenzymatic Synthesis of Sulfated and Nonsulfated Core 2 O-GalNAc Glycans

Zhuojia Xu et al.

Summary: A diversity-oriented chemoenzymatic approach was used for the collective preparation of sulfated core 2 O-GalNAc glycans and their nonsulfated counterparts. A sulfated trisaccharide and a nonsulfated trisaccharide were chemically synthesized, then enzymatic extension was carried out using a panel of robust glycosyltransferases to achieve regioselective sialylation, resulting in the generation of 36 structurally well-defined O-GalNAc glycans.

A facile chemoenzymatic synthesis of SARS-CoV-2 glycopeptides for probing glycosylation functions

Guanghui Zong et al.

Summary: Glycosylation is crucial in SARS-CoV-2 infection. A new facile chemoenzymatic synthesis method was used to create core-fucosylated N-glycopeptides derived from the SARS-CoV-2 Spike protein, which were then tested for binding with glycan-dependent neutralizing antibody S309 and human lectin CLEC4G. The synthetic glycopeptides serve as valuable tools for further characterization of viral glycosylation.
