Implications of statin metabolism-related gene testing in guiding individualized drug use in cardiovascular diseases

Yiliang Zhang et al.

Summary: The purpose of this study was to determine the value of statin metabolism-related gene testing in guiding individualized drug use in cardiovascular and cerebrovascular diseases. A total of 158 eligible patients were recruited and assigned to different groups based on their diseases. The results showed significant differences in the effectiveness of statins in patients with different genotypes, suggesting that genetic testing is valuable in guiding medication use in patients with cardiovascular disease.

A Brain-Penetrant and Bioavailable Pyrazolopiperazine BACE1 Inhibitor Elicits Sustained Reduction of Amyloid β In Vivo

Aldo Peschiulli et al.

Summary: This study describes further work on prototypes of heteroaryl-fused piperazine inhibitors of BACE1, aiming to modulate their basicity and reduce binding to the hERG channel. The researchers identified compound 36, which demonstrated nanomolar potency, cardiovascular safety, and oral bioavailability, and effectively reduced A beta(42) levels in animal models.

Faulty autolysosome acidification in Alzheimer's disease mouse models induces autophagic build-up of Aβ in neurons, yielding senile plaques

Ju-Hyun Lee et al.

Summary: This study reveals unique autophagy dysregulation within neurons in Alzheimer's disease mouse models, particularly deficient autolysosome acidification, leading to the accumulation of autophagic vacuoles and the formation of flower-like structures called "PANTHOS," which may contribute to the development of senile plaques in Alzheimer's disease.

Dual-target compounds for Alzheimer's disease: Natural and synthetic AChE and BACE-1 dual-inhibitors and their structure-activity relationship (SAR)

Joao P. S. Ferreira et al.

Summary: Alzheimer's disease (AD) is a major cause of dementia worldwide, with no disease-modifying treatments currently available. Distinct pharmacological targets like Acetylcholinesterase (AChE) and B-secretase-1 (BACE-1) have been explored for AD treatment. Research is focused on developing multi-target directed ligands as an alternative approach to single-target drugs.

Design, Synthesis and Evaluation of 2,4,6-substituted Pyrimidine Derivatives as BACE-1 inhibitor: Plausible Lead for Alzheimer's Disease

Priti Jain et al.

Summary: Alzheimer's disease is a common neurodegenerative disorder, with BACE-1 playing a key role in its pathogenesis. This study focused on designing small molecular weight compounds to inhibit BACE-1, which showed strong interaction with the active sites of BACE-1 and good oral bioavailability and brain permeability. The compounds displayed potent BACE-1 inhibition in vitro, with a correlation between docking score and activity.

A review on ferulic acid and analogs based scaffolds for the management of Alzheimer's disease

Yash Pal Singh et al.

Summary: Alzheimer's disease is an age-related neurodegenerative disorder characterized by central cholinergic neuronal loss leading to cognitive dysfunction and impaired motor activity. Current therapies only offer symptomatic relief without addressing the underlying factors, and there is no drug to slow down disease progression. Researchers are investigating new hybrid compounds, such as ferulic acid-based derivatives, as potential treatments for Alzheimer's disease.
