Modulatory Properties of Food and Nutraceutical Components Targeting NLRP3 Inflammasome Activation

Mattia Spano et al.

Summary: Inflammasomes are key proteins that initiate cellular inflammatory signaling pathway. NLRP3 inflammasome is one of the main protein complexes involved in inflammatory events. Recent studies show that dietary intake and nutritional status can modulate metabolic inflammation, and various bioactive compounds found in plant foods have been shown to possess anti-NLRP3 inflammasome activity.

Differential susceptibility to lipopolysaccharide affects the activation of toll-like-receptor 4 signaling in THP-1 cells and PMA-differentiated THP-1 cells

Young Kyu Kim et al.

Summary: The study found that pro-inflammatory cytokines were induced to a greater extent in PMA-differentiated THP-1 cells compared to THP-1 cells, due to the higher preservation of TLR4-related proteins and intracellular adaptor proteins. MyD88 was identified as a key molecule affecting the difference in LPS susceptibility, and molecules related to the MAPK and NF-kappa B pathways showed higher levels of phosphorylation in PMA-differentiated THP-1 cells.

Pharmacokinetics-On-a-Chip: In Vitro Microphysiological Models for Emulating of Drugs ADME

Qasem Ramadan et al.

Summary: Despite the ongoing efforts in pharmaceutical and biotech industries, drug development remains costly and high-risk, with debates over the use of animal models due to ethical concerns and pathophysiological differences between animals and humans. Current in vitro models lack the functionality to fully replace animal models, but the development of micro-physiological systems shows potential as tools for predicting human pharmacokinetics and pharmacodynamics. The integration of ADME processes in vitro is a significant development meeting important pharmaceutical industry needs.

EPA and DHA differentially modulate monocyte inflammatory response in subjects with chronic inflammation in part via plasma specialized pro-resolving lipid mediators: A randomized, double-blind, crossover study

Jisun So et al.

Summary: The study found that EPA and DHA have different effects on monocyte inflammatory response induced by chronic inflammation, with DHA showing a broader role in inhibiting pro-inflammatory cytokines. These differential effects may be mediated by different groups of PUFA derivatives, demonstrating the immunomodulatory activities of SPM and their intermediates.

Curcumin steers THP-1 cells under LPS and mTORC1 challenges toward phenotypically resting, low cytokine-producing macrophages

Harleen Kaur et al.

Summary: The study demonstrated that curcumin and its combination with piperine can suppress mTORC1 kinase activity, reduce inflammatory response of THP-1 macrophages, and inhibit M1 and M2 polarization signaling pathways. Curcumin also modulated monocyte differentiation into M0 macrophage phenotype with low inflammatory cytokine output.

In vitro assessment of the cytotoxicity and anti-inflammatory properties of a novel dietary supplement

Ionela D. Popescu et al.

Summary: Studies have shown that certain plant extracts have anti-inflammatory potential and can reduce inflammatory processes and related diseases. This study demonstrated the excellent performance of a novel lingonberry-based dietary supplement in suppressing the inflammatory response without cytotoxicity in monocytes/macrophages.

Overconsumption of Omega-6 Polyunsaturated Fatty Acids (PUFAs) versus Deficiency of Omega-3 PUFAs in Modern-Day Diets: The Disturbing Factor for Their Balanced Antagonistic Metabolic Functions in the Human Body

Abeba Haile Mariamenatu et al.

Summary: Polyunsaturated fatty acids, specifically Omega-3 and Omega-6, play crucial roles in human health, with an imbalance in their intake ratio potentially leading to inflammations and vascular constriction. To mitigate these risks, it is recommended to consume a balanced ratio of Omega-6 and Omega-3 at 4-5/1.
