Manganese-phenolic nanoadjuvant combines sonodynamic therapy with cGAS-STING activation for enhanced cancer immunotherapy

Hao Tian et al.

Summary: A phenolic nanoadjuvant has been developed to enhance the maturation of dendritic cells (DCs) and improve the efficacy of cancer immunotherapy. This nanoadjuvant promotes the activation of the cGAS-STING pathway and sensitizes tumors to PD-L1 checkpoint blockade immunotherapy, resulting in inhibiting tumor growth and restraining lung metastasis.

cGAS-STING signaling in cancer immunity and immunotherapy

Huashan Du et al.

Summary: Recent studies have revealed that the cGAS-STING pathway plays a crucial role in antitumor immunity, promoting senescence and apoptosis of cancer cells while enhancing the protective effects of cytotoxic T cells and natural killer cells. Combining the cGAS-STING signaling pathway with other therapeutic methods offers a new perspective for overcoming obstacles in its application.

Maintaining manganese in tumor to activate cGAS-STING pathway evokes a robust abscopal anti-tumor effect

Chao Wang et al.

Summary: Synchronizing Mn2+ delivery with accumulated cytosolic DNA after radiotherapy can promote the activation of the cGAS-STING pathway and enhance antitumor immunity. This strategy can achieve over 90% inhibition rate even in unirradiated tumors, providing a simple and novel approach to effectively activate the immune response in tumors.

Summary: SAVI is a type I interferonopathy caused by gain-of-function mutations in the STING1 gene, characterized by systemic inflammation, skin vasculopathy, and ILD. Patients usually present with early symptoms, and the severity of ILD varies. Tissue loss and T-cell defects are common features.

Manganese salts function as potent adjuvants

Rui Zhang et al.

Summary: The colloidal manganese salt (Mn jelly, MnJ) acts as both an immune potentiator and a delivery system, promoting immune responses by inducing antibody production and CD4(+)/CD8(+) T-cell proliferation and activation. It also shows promise as a mucosal adjuvant, inducing high levels of secretory IgA and proving effective for various antigens, making it a potential candidate for adjuvant use.

NLRP3 inflammasome activation and cell death

Yi Huang et al.

Summary: The NLRP3 inflammasome is a cytosolic multiprotein complex that responds to microbial infection, endogenous danger signals, and environmental stimuli. Its activation can induce pyroptosis and release of inflammatory cytokines, contributing to innate immune defense. However, aberrant activation of the NLRP3 inflammasome is associated with inflammatory diseases, and there is a close relationship between cell death pathways and NLRP3 inflammasome activation.
