Selective Targeting of Cancer Cells by Copper Ionophores: An Overview

Valentina Oliveri

Summary: Conventional cancer therapies face challenges due to off-target effects, leading to the need for new therapeutic agents with increased selectivity, reduced side effects, and the ability to overcome cancer cell resistance targeting cancer stem cells. Some copper ionophores show promise in inducing cuproptosis in cancer cells with greater selectivity.

Disulfiram and metformin combination anticancer effect reversible partly by antioxidant nitroglycerin and completely by NF-κB activator mebendazole in hamster fibrosarcoma

Kosta J. Popovic et al.

Summary: The combination of disulfiram and metformin showed significant inhibition of fibrosarcoma growth in hamsters without toxicity, while single treatments did not have a significant anti-tumor effect. Nitroglycerin partially rescued tumor progression, possibly through ROS inhibition, while mebendazole completely blocked the anticancer activity of the disulfiram and metformin combination.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Anti-Cancer Effects of Zotarolimus Combined with 5-Fluorouracil Treatment in HCT-116 Colorectal Cancer-Bearing BALB/c Nude Mice

Geng-Ruei Chang et al.

Summary: The study demonstrates that Zotarolimus alone and in combination with 5-FU can inhibit the growth and metastasis of colorectal adenocarcinoma, as well as increase the sensitivity of tumor cells to chemotherapy. Additionally, the combination therapy shows a synergistic effect in retarding tumor growth and enhancing tumor inhibition. These findings suggest that Zotarolimus may serve as a potential anti-tumor agent in the treatment of human colon adenocarcinoma, with potential for further therapeutic development.

Recent Advances in Repurposing Disulfiram and Disulfiram Derivatives as Copper-Dependent Anticancer Agents

Vinodh Kannappan et al.

Summary: Copper plays a key role in cancer progression and copper-based complexes are being investigated as novel anticancer drugs. The combination of DSF with copper has shown great anticancer potential, but DSF has proven unsuccessful in clinical trials, likely due to poor stability of the currently used oral version of DSF.
