Conquering the Hypoxia Limitation for Photodynamic Therapy

Summary: Photodynamic therapy (PDT) is a promising treatment modality due to its high spatiotemporal selectivity, minimal invasiveness, and low systemic toxicity, however, the hypoxic nature of many solid tumors limits its therapeutic effect. Efforts have been made to increase tumor oxygen content and alleviate tumor hypoxia through various strategies developed in the past decade, including delivering exogenous oxygen, generating oxygen in situ, reducing tumor cellular oxygen consumption, regulating the tumor microenvironment, and inhibiting the hypoxia-inducible factor 1 signaling pathway.

Enhanced chemo-photodynamic therapy of an enzyme-responsive prodrug in bladder cancer patient-derived xenograft models

Summary: Patient-derived xenograft (PDX) models are powerful tools for cancer biology and drug discovery. A polymeric nano-sized drug delivery system, composed of a photosensitizer and a cathepsin B-sensitive polymer-paclitaxel prodrug, showed great tumor targeting and anti-tumor effects in human bladder cancer PDX models. Combinational chemotherapy and PDT resulted in up-regulation of oxidative phosphorylation and ROS generation, while down-regulating pathways related to tumor progression, invasion, and metastasis, making it a potential candidate for bladder cancer therapy.

Summary: The limited therapeutic efficacy of photodynamic therapy (PDT) to hypoxic solid tumors has been improved by two new benzothiophenylisoquinoline-derived cyclometalated Ir-III complexes, IrL1 and MitoIrL2. Both complexes demonstrated a type I PDT process and induced photoinduced ferroptosis in tumor cells under hypoxia, with MitoIrL2 outperforming IrL1 in inhibiting tumor growth through a synergistic effect of ferroptosis and apoptosis.

Amorphous silica nanoparticles induce inflammation via activation of NLRP3 inflammasome and HMGB1/TLR4/MYD88/NF-kb signaling pathway in HUVEC cells

Summary: The study revealed that amorphous SiO2 NPs induce pro-inflammatory responses in HUVECs by regulating signal pathways, leading to increased levels of ROS, LDH, inflammatory factors, and cell adhesion molecules. The translocation and release of HMGB1, regulated by ROS and NLRP3 inflammasome, was shown to play a key role in activating the NF-kappa B signaling pathway and inducing the inflammatory response in HUVECs. This provides new insights into the cytotoxicity induced by SiO2 NPs and offers safety guidance for further nanomaterial applications.

Pyroptosis at the forefront of anticancer immunity

Summary: Pyroptosis, a lytic and inflammatory programmed cell death pathway, has shown potential in addressing tumor resistance to apoptosis and the immunosuppressive tumor microenvironment by inducing a robust inflammatory response and marked tumor regression. Gasdermin proteins mediate pyroptosis and facilitate immune cell activation and infiltration, but aberrant pyroptosis may also contribute to tumor progression by upregulating gasdermin proteins in certain cancers. Harnessing pyroptosis as a tool in anticancer strategies shows promise in novel interventions.

The relationship between expression of PD-L1 and HIF-1α in glioma cells under hypoxia

Summary: The study showed a positive correlation between PD-L1 and HIF-1α in glioma, with their overexpression associated with poor survival. HIF-1α was found to induce PD-L1 expression under hypoxic conditions, which could be abrogated by inhibiting HIF-1α.
