Summary: In this study, we successfully fabricated metformin-based supramolecular nanodrugs with an A beta-responsive on-demand drug release for synergistic Alzheimer's disease therapy. The nanodrugs exhibited outstanding physiological stability and selectively penetrated the blood-brain barrier to target microglia and neurons. The released drugs, metformin and donepezil, showed a superior synergistic anti-dementia effect by enhancing microglial phagocytosis and A beta clearance.

Furthermore, hGLV loaded with photothermal agent achieved excellent photothermal therapy (PTT) in eliminating tumors, inducing immunogenic cell death, generating tumor-associated antigens, and promoting immune responses with the help of co-encapsulated immune adjuvant. Such hybrid nanovesicles show promise in drug delivery for cancer treatment.
