Abnormal Level of Manganese, Iron, Iodine, and Selenium in the Hair of Children Living in Kashin-Beck Disease Endemic Areas

Feng'e Zhang et al.

Summary: This study analyzed the bio-element profile of Kashin-Beck disease (KBD) and found that manganese, iron, and iodine (in conjunction with selenium) play a vital role in the etiology and pathogenesis of KBD. High manganese and iron levels with low selenium and iodine levels are characteristic features of the bio-element profile of KBD.

An eQTL variant of ALDH1A2 is associated with Kashin-Beck disease in Chinese population

Di Zhang et al.

Summary: This study found a close association between Kashin-Beck disease (KBD) and the ALDH1A2 gene variant rs3204689, which is linked to the high expression of ALDH1A2. ALDH1A2 was up-regulated in the cartilage and whole blood of KBD patients and may play a role in the pathogenesis of KBD.

Investigation of selenium nutritional status and dietary pattern among children in Kashin-Beck disease endemic areas in Shaanxi Province, China using duplicate portion sampling method

Yujie Ning et al.

Summary: This study aimed to investigate the selenium intake of children in endemic areas after termination of selenium supplementation, and evaluate their nutritional status and dietary structure. The results showed that the selenium levels and dietary selenium intake of children in endemic areas were significantly lower than those in non-endemic areas. The dietary structure of children in endemic areas was undiversified, leading to limited intake of multiple nutrients.

The Status of Selenium and Zinc in the Urine of Children From Endemic Areas of Kashin-Beck Disease Over Three Consecutive Years

Xin Kang et al.

Summary: This study aimed to detect the status of selenium and zinc in the urine of children from endemic areas of Kashin-Beck disease (KBD) over three consecutive years. The results showed that the selenium levels in children from KBD endemic areas were lower than normal, indicating the need for constant monitoring and adjustment of selenium supplementation.

Upregulated expression of transforming growth factor-β receptor I/II in an endemic Osteoarthropathy in China

Ying Zhang et al.

Summary: This study evaluated the levels of TGF-beta receptor I and II in clinical samples of KBD and in pre-clinical disease models. The results suggest that TGF-beta RI and II may be involved in the pathophysiology of KBD, providing new insights into the pathways that contribute to KBD development.

Pleiotropic Roles of NOTCH1 Signaling in the Loss of Maturational Arrest of Human Osteoarthritic Chondrocytes

Manuela Minguzzi et al.

Summary: Notch signaling pathway plays a critical role in cartilage development and homeostasis, with NOTCH1 being the key receptor that can affect the health status of osteoarthritis chondrocytes by regulating proliferation, hypertrophy, and terminal differentiation. Silencing NOTCH1 showed a healthier phenotype in OA chondrocytes, suggesting it is a convenient therapeutic target to attenuate OA progression.

Relevance of Notch Signaling for Bone Metabolism and Regeneration

Tobias M. Ballhause et al.

Summary: Notch1-4 receptors and their signaling pathways are crucial in cell fate decision by coordinating cell proliferation, differentiation and apoptosis, with differential expression and activation observed in various organs and tissues. In bone, Notch receptors control remodeling and regeneration, with distinct expression and function depending on differentiation stages. The crosstalk between Notch signaling and other pathways in bone turnover, including Wnt/beta-catenin, BMP and RANKL/OPG, is outlined.
