The influence of FAAH genetic variation on physiological, cognitive, and neural signatures of fear acquisition and extinction learning in women with PTSD

Kevin M. Crombie et al.

Summary: This study found that genetic variation in the FAAH gene influences the physiological, cognitive, and neural fear learning characteristics in women with PTSD. Different genotypes showed divergent physiological and neural responses, suggesting that targeting FAAH may be a promising therapeutic approach for PTSD.

Enhancing exposure therapy for posttraumatic stress disorder (PTSD): a randomized clinical trial of virtual reality and imaginal exposure with a cognitive enhancer

JoAnn Difede et al.

Summary: Posttraumatic stress disorder (PTSD) is a significant public health issue with limited treatment options. This study tested the efficacy of virtual reality exposure (VRE) or prolonged imaginal exposure (PE) augmented with D-cycloserine (DCS) for combat-related PTSD, and explored the potential moderating effects of brain-derived neurotrophic factor (BDNF) and fatty acid amide hydrolase (FAAH). The results showed similar clinical improvement for both VRE and PE, with VRE being more effective for depressed participants and PE being more effective for nondepressed participants. The study also found a potential moderating effect of genetic markers on treatment response.

Fatty acid amide hydrolase is lower in young cannabis users

Maya R. Jacobson et al.

Summary: Lower levels of FAAH enzyme were found in the brains of adult cannabis users during early abstinence, with higher cannabis use throughout the past year correlating with decreased [C-11]CURB binding. The study also detected lower [C-11]CURB binding in a younger cohort with less cumulative cannabis exposure, confirming and extending previous findings.

Hypothalamic endocannabinoids in obesity: an old story with new challenges

Cristina Miralpeix et al.

Summary: The hypothalamus plays a crucial role in the pathogenesis of obesity, with disrupted endocannabinoid system considered a landmark indicator. Recent research has expanded knowledge of the complex endocannabinoid system, focusing on CB receptors, ligands, non-CB receptors, and metabolizing enzymes, particularly in the hypothalamus. The insights gained have provided promising targets for tackling obesity and related diseases.

Summary: Individuals with ASPD have lower FAAH density in the amygdala, while FAAH expression in the cerebellum and striatum is inversely related to impulsivity. Cerebellar FAAH density is also negatively associated with assaultive aggression.

The Endocannabinoid Anandamide Attenuates Acute Respiratory Distress Syndrome by Downregulating miRNA that Target Inflammatory Pathways

Muthanna Sultan et al.

Summary: This study demonstrates that endocannabinoids like AEA can alleviate ARDS induced by SEB by suppressing inflammation through down-regulation of key miRNA that regulate immunosuppressive pathways. Treatment with AEA in mice showed improvements in lung function, reduced inflammatory cell infiltrate, and modulation of pro-inflammatory cytokines and immune cells in the lungs. Additionally, AEA altered miRNA expression, leading to the induction of anti-inflammatory phenotypes and the regulation of immunosuppressive pathways.
