Chordoma A Review and Differential Diagnosis

Veronica Ulici et al.

Summary: Chordoma is a clinically and histologically unique malignant neoplasm that requires the exclusion of many diagnostic considerations to establish the correct diagnosis. Treatment options have primarily focused on surgical excision with limited success; however, novel therapeutic options including targeted therapy and immunotherapy hold promise for improving prognosis.

Definitive high-dose, proton-based radiation for unresected mobile spine and sacral chordomas

Walter Banfield et al.

Summary: This study retrospectively reviewed the outcomes of high-dose proton-based definitive radiotherapy for newly diagnosed, unresected spinal and sacral chordomas. The results showed that high-dose definitive radiotherapy is an effective treatment option for medically inoperable or otherwise unresected mobile spine or sacrococcygeal chordomas.

Aldehyde Dehydrogenase, a Therapeutic Target in Chordoma: Analysis in 3D Cellular Models

Marie-Anais Locquet et al.

Summary: Chordomas are rare, slow-growing tumors of the axial skeleton that are locally aggressive and refractory to conventional therapies. Radical surgery and radiation remain the first-line treatments, but inhibiting ALDH has been shown to increase radiosensitivity in chordoma 3D models. Further research in this direction could lead to new treatment options for chordoma patients.

Patient-Derived Cancer Organoids as Predictors of Treatment Response

Maikel Verduin et al.

Summary: Patient-derived cancer organoids are valuable tools in predicting individualized treatment responses, comparing treatment efficacies, and screening new anti-cancer drugs. However, further improvements are needed to fully utilize the potential of cancer organoids in clinical cancer therapy management.
