A modular map of Bradykinin-mediated inflammatory signaling network

D. A. B. Rex et al.

Summary: Bradykinin signals through its receptor B2R with diverse vascular permeability functions and is implicated in inflammation, chronic pain, vasculopathy, neuropathy, obesity, diabetes, and cancer. An integrated pathway reaction map of bradykinin consisting of 233 reactions has been developed, which will aid in identifying novel therapeutic targets for diseases associated with dysregulated bradykinin signaling.

Mesenchymal stem cell-glioblastoma interactions mediated via kinin receptors unveiled by cytometry

Micheli Mainardi Pillat et al.

Summary: Glioblastoma (GBM) is a highly malignant and therapy-resistant brain tumor characterized by rapid invasion and interactions with stromal cells. Recent evidence highlights interactions between GBM cells and mesenchymal stem cells (MSCs) through direct cell-cell contact or fusion. Paracrine interactions involve cytokine signaling, regulating MSC tropism towards GBM and immune responses, with MSC effects critical for tumor progression. Kinins and kinin-B1 and -B2 receptors facilitate information flow between MSCs and GBM cells, influencing tumor migration and fusion.

The transformation of cancer-associated fibroblasts: Current perspectives on the role of TGF-β in CAF mediated tumor progression and therapeutic resistance

Bikash Chandra Jena et al.

Summary: TGF-beta is considered as a crucial mediator of cell growth and cancer cell progression through both SMAD-dependent and independent pathways. In the tumor microenvironment, cancer-associated fibroblasts (CAFs) play a prominent role in tumor progression and chemoresistance, with TGF-beta facilitating the crosstalk between CAFs and tumor cells. Targeting the TGF-beta and CAFs interaction in clinical trials is essential for developing successful anti-cancer therapies.

Acid-responsive HPMA copolymer-bradykinin conjugate enhances tumor-targeted delivery of nanomedicine

Enoch Appiah et al.

Summary: This study designed and synthesized a novel HPMA-based copolymer conjugate of BK, which showed tumor-selective action and improved accumulation of nanomedicines in the tumor, resulted in enhanced intratumoral blood flow and increased tumor deposition of nanomedicines.

Novel Bradykinin Receptor Inhibitors Inhibit Proliferation and Promote the Apoptosis of Hepatocellular Carcinoma Cells by Inhibiting the ERK Pathway

Yiou Wang et al.

Summary: The novel BK receptor inhibitors J051-71 and J051-105 were found to reduce the viability of liver cancer cells and inhibit colony formation, possibly by inhibiting the ERK pathway. These inhibitors also reversed the cell proliferation induced by BK by downregulating B1 receptor expression, suggesting a potential target for HCC therapy.
