Adaptive chromatin remodeling and transcriptional changes of the functional kinome in tumor cells in response to targeted kinase inhibition

Michael P. East et al.

Summary: Pharmacological inhibition of protein kinases alters gene expression and induces adaptive reprogramming of tumor cell regulatory networks. This reprogramming is dependent on changes in open chromatin state and the activity of specific transcription factors. The dynamic plasticity of protein kinase expression is correlated with adaptive resistance to targeted kinase inhibition.

Multivalent DNA and nucleosome acidic patch interactions specify VRK1 mitotic localization and activity

Gabrielle R. Budziszewski et al.

Summary: The chromatin kinase VRK1 interacts with both linker DNA and the nucleosome acidic patch to phosphorylate H3T3. Mutations in VRK1 that interfere with acidic patch binding lead to mislocalization of VRK1 during mitosis, potentially providing a new molecular mechanism for pathogenesis.

Adult-Onset Spinal Muscular Atrophy due to Mutations in the VRK1 Gene

Angela Sung et al.

Summary: This study aimed to explore clinical phenotypes associated with VRK1 gene mutations. Two individuals with slowly progressive weakness and symptoms consistent with adult-onset spinal muscular atrophy without pontocerebellar atrophy were found to have likely pathogenic variants in the VRK1 gene, expanding the clinical spectrum of neurologic disorders linked to VRK1 mutations. Both patients in the study were of Hispanic descent.

Summary: This study conducted genetic and molecular functional analysis on a family with hereditary spastic paraplegia (HSP) affected members and identified a novel pathogenic VRK1 variant associated with a neurologic phenotype. The VRK1 variant impairs the formation of CBs and DNA damage response in patients with HSP.

The RNF8 and RNF168 Ubiquitin Ligases Regulate Pro- and Anti-Resection Activities at Broken DNA Ends During Non-Homologous End Joining

Bo-Ruei Chen et al.

Summary: RNF8 and RNF168 play crucial roles in non-homologous end joining (NHEJ) repair by recruiting 53BP1 to protect DNA ends from nucleases, and they regulate pathways that both prevent and promote DNA end resection in cells arrested in G0/G1 phase.

The human VRK1 chromatin kinase in cancer biology

Ignacio Campillo-Marcos et al.

Summary: VRK1 is a nuclear Ser-Thr chromatin kinase that is not mutated in cancer, but is overexpressed in many types of tumors and linked to poor prognosis. It plays crucial roles in regulating cell proliferation and DNA damage response pathways, having both pro-tumorigenic effects and protective anti-tumor roles depending on the cellular context.

Summary: The deregulation of chromatin modification underlies numerous oncogenic processes, contributing to cancer development by inducing epigenetic, transcriptomic, and phenotypic alterations. Understanding and developing more effective drugs to target these chromatin-related vulnerabilities will enhance the treatment of human malignancies. This review synthesizes the connections between chromatin modifications and cancer, highlighting options for therapeutic targeting based on recent advances.
