Purification of functional Plasmodium falciparum tubulin allows for the identification of parasite-specific microtubule inhibitors

Summary: Cytoskeletal proteins are crucial for parasite survival and the study of parasite tubulin may lead to the development of novel antimalarials. This study successfully purified and characterized tubulin from Plasmodium falciparum, finding that it shares similarity with plant tubulin rather than mammalian tubulin. This opens up the possibility of finding compounds that can selectively disrupt parasite microtubules without affecting the host cell cytoskeleton.

GTP-dependent formation of straight tubulin oligomers leads to microtubule nucleation

Summary: This study demonstrates that the formation of straight oligomers of critical size is essential for nucleation of microtubules. Both GDP and GTP tubulin form single-stranded oligomers with different curvature distributions, and the proportion of straight oligomers can accelerate nucleation. The results suggest that cellular factors involved in nucleation promote it via stabilization of straight oligomers.

Gliding motility of Plasmodium merozoites

Summary: Plasmodium merozoites, traditionally not known to undergo gliding motility, were discovered to possess this ability in vitro, which may be crucial for successful parasite invasion. The distinct merozoite motility profiles of human infective species, Plasmodium falciparum and Plasmodium knowlesi, suggest different invasion strategies. The development of a higher throughput assay to study genetic and pharmacological perturbations on merozoite motility provides a new avenue for research on treatments targeting blood stage invasion pathways.
