Determination of dissociation constants of protein ligands by thermal shift assay

Jaina A. Bhayani et al.

Summary: The thermal shift assay is a powerful tool for detecting molecular interactions between proteins and ligands by tracking protein unfolding using temperature and an extrinsic fluorescent dye. A new analysis method based on equilibrium systems and the van't Hoff equation is proposed for determining Kd, showing promising results in testing with different proteins and enzymes. This method has the potential to offer broad applicability across protein classes and detect both active and allosteric site binding compounds.

Coconut Mesocarp-Based Lignocellulosic Waste as a Substrate for Cellulase Production from High Promising Multienzyme-Producing Bacillus amyloliquefaciens FW2 without Pretreatments

Van Hong Thi Pham et al.

Summary: The study aimed to investigate cellulase production from coconut-mesocarp by the bacterial strain FW2 isolated from kitchen food waste in South Korea. The strain exhibited tolerance to a wide range of temperature, pH, and high salt concentrations. Optimal enzyme activity conditions were found to be 40-50 degrees C, pH 7.0-7.5, and 0-10% NaCl, resulting in high levels of CMCase, Avicelase, and FPase activities. The results suggest that enzyme production by strain FW2 may have significant commercial values for various applications including industry and waste treatment.

Pathways of lignocellulosic biomass deconstruction for biofuel and value-added products production

Shams Forruque Ahmed et al.

Summary: This paper addresses the gaps in the existing literature review works by comprehensively evaluating different pretreatment methods and applications of lignocellulosic biomass (LCB) and incorporating techno-economic considerations. The review finds that physical and chemical pretreatment methods, although effective, are highly dependent on energy and chemicals, producing waste and inhibitory compounds. Biological pretreatment can address these challenges but has longer incubation times. Integrated/coupling methods have gained popularity for improving chemical yield but come at a high cost of operation. Process integration and optimization are needed to make pretreatment processes more techno-economically feasible.
