Effects of orphan histidine kinases on clostridial sporulation progression and metabolism

Guangqing Du et al.

Summary: The study explored the regulatory roles of histidine kinases (HKs) in clostridial differentiation, highlighting the potential of HK engineering to control sporulation and enhance butanol biosynthesis. The comparative genomic analysis identified the importance of cac3319 and cac0437 in regulating sporulation and acids and butanol biosynthesis. Morphological analysis showed that engineering HKs altered the progression of sporulation, with cac3319 knockout abolishing sporulation and cac0437 knockout initiating but blocking sporulation before Stage II.

Role of ARK5 in cancer and other diseases (Review)

Guoheng Mo et al.

Summary: ARK5, a member of the AMPK family, plays a critical role in energy regulation and hypoxia. Overexpression of ARK5 is associated with tumor invasion, metastasis, and cancer cell malignancy, with potential as a therapeutic target in cancer treatment. Furthermore, ARK5 is also involved in various other diseases and physiological processes, suggesting its broad impact beyond cancer.

Phosphoproteome Study of Escherichia coli Devoid of Ser/Thr Kinase YeaG During the Metabolic Shift From Glucose to Malate

Abida Sultan et al.

Summary: Understanding phosphorylation-mediated regulation of metabolic enzymes and pathways during metabolic shifts is a major challenge. In this study, proteome and phosphoproteome analysis of E. coli Delta yeaG revealed the phosphorylation of AceA by YeaG during the shift from glucose to malate, suggesting a role in this metabolic transition. SILAC-based approaches have shown utility in identifying kinase substrates under relevant physiological conditions.

The DEAD-box RNA helicase RhIE2 is a global regulator of Pseudomonas aeruginosa lifestyle and pathogenesis

Stephane Hausmann et al.

Summary: The study reveals the functional and molecular divergence between two homologous RNA helicases, RhIE1 and RhIE2, which contribute to bacterial fitness and pathogenesis. RhIE1 is mainly required for growth in the cold, while RhIE2 acts as a global post-transcriptional regulator affecting the levels of hundreds of cellular transcripts. The global impact of RhIE2 is mediated by its unique C-terminal extension.

Ser/Thr Kinase-Dependent Phosphorylation of the Peptidoglycan Hydrolase CwlA Controls Its Export and Modulates Cell Division in Clostridioides difficile

Transito Garcia-Garcia et al.

Summary: The STK signaling pathway regulates PGH homeostasis to precisely control PG hydrolysis during cell division. The study on PG hydrolase CwlA in C. difficile reveals its important role in cell division and its regulation by PrkC, highlighting a novel regulatory mechanism by which STK controls protein export.

Energy-efficient butanol production by Clostridium acetobutylicum with histidine kinase knockouts to improve strain tolerance and process robustness

Guangqing Du et al.

Summary: This study identified four orphan histidine kinases (HKs) that control sporulation and solventogenesis in Clostridium acetobutylicum. Single knockout of cac3319 halted sporulation at the initiation stage but increased butanol tolerance and production, while cac0437 knockout caused premature sporulation and inhibited the transition to solventogenesis, resulting in low butanol production.
