Biochemical characterization and cleavage pattern analysis of a novel chitosanase with cellulase activity

Summary: In this study, a novel GH5 chitosanase OUC-Csngly with dual chitosanase and cellulase activities was cloned and characterized. Molecular docking analysis revealed that the C-2 group of sugar units influenced the binding of the enzyme to oligosaccharides, resulting in different cleavage patterns. Additionally, OUC-Csngly exhibited distinctive cleavage patterns towards two different types of oligosaccharides.

Summary: In this study, a chitosanase gene was cloned and optimized to obtain a recombinant strain with high activity for chitosan oligosaccharide preparation. The purified chitosanase showed stability and high activity towards colloidal chitosan, and could control the production of chitosan oligosaccharides with different degrees of polymerization.

Gene Cloning, Functional Expression, and Characterization of a Novel GH46 Chitosanase from Streptomyces avermitilis (SaCsn46A)

Summary: The novel glycoside hydrolase family 46 chitosanase SaCsn46A from Streptomyces avermitilis has been successfully cloned and functionally expressed. This enzyme shows high stability and activity in moderate temperature and pH conditions, with the ability to hydrolyze a variety of polysaccharides linked by beta-1,4-glycosidic bonds. Moreover, it can be enhanced by divalent metal ions like Mg2+ and Mn2+, making it a promising candidate for producing glucosamine and chitooligosaccharides from chitosan.

New Insights into Bifunctional Chitosanases with Hydrolysis Activity toward Chito- and Cello-Substrates

Summary: In this study, we investigated the bifunctional activity of GH46 chitosanases based on cleavage specificity classification. We found that CsnMHK1 from Bacillus circulans MH-K1, previously thought to only hydrolyze chitosan, can also hydrolyze cello-substrates. Comparison with GH8 chitosanase provided new insights into substrate specificities and challenged the current understanding of chitosanase classification. These findings contribute to our understanding of chitosanase recognition and hydrolysis, which is valuable for future studies.

New Class of Chitosanase from Bacillus amyloliquefaciens for the Generation of Chitooligosaccharides

Summary: Chitooligosaccharides (COS), derived from chitin or chitosan, have diverse applications. A chitosanase from Bacillus amyloliquefaciens (BamCsn) was characterized, with the crucial role of a tryptophan residue, W204, identified for its activity. BamCsn exhibited unique properties such as transglycosylation and the ability to cleave chitin oligosaccharides, showing potential for COS generation.

Preparation of Active Chitooligosaccharides with a Novel Chitosanase AqCoA and Their Application in Fungal Disease Protection

Summary: A newly identified endo-chitosanase AqCoA showed promising antifungal activity by inhibiting appressorium formation of Magnaporthe oryzae and inhibiting the growth of both M. oryzae and Fusarium oxysporum. The resulting chitooligosaccharides also acted as immune elicitors in rice, demonstrating the potential of AqCoA and its products as effective tools for plant protection against pathogenic fungi.

Machine learning reveals sequence-function relationships in family 7 glycoside hydrolases

Summary: GH7 family glycoside hydrolases are key enzymes in cellulose degradation, including cellobiohydrolases and endoglucanases. Machine-learning models show that the number of residues in the active-site loops strongly correlates with functional subtype across the GH7 family, with specific residues at certain sequence positions accurately predicting the subtype. Additionally, a random forest model trained on residues in the catalytic domain can predict the presence of a carbohydrate-binding module with high accuracy.
