Copper induces cell death by targeting lipoylated TCA cycle proteins

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

The TGF-β/HDAC7 axis suppresses TCA cycle metabolism in renal cancer

Summary: Evidence suggests alterations in mitochondrial metabolism in renal cell carcinoma, specifically in the regulation of the TCA cycle. Reduced TCA cycle enzyme expression, notably in correlation with transcription factor PGC-1 alpha loss, is more pronounced in RCC compared to other tumor types. Mechanistically, TGF-beta signaling, in partnership with HDAC7, suppresses TCA cycle enzyme expression, and pharmacologic inhibition of TGF-beta restores enzyme expression and suppresses tumor growth.

Comprehensive Analysis of Ferroptosis Regulators With Regard to PD-L1 and Immune Infiltration in Clear Cell Renal Cell Carcinoma

Summary: Our study revealed that ferroptosis regulators are correlated with the immune-related gene PD-L1 in clear cell renal cell carcinoma (ccRCC). Consensus clustering analysis identified two subtypes, with subtype 1 showing better prognosis and higher PD-L1 expression, while subtype 2 exhibited higher levels of immune cell infiltration. Enrichment analysis indicated significant pathways in subtype 1, suggesting their important role in ccRCC progression.
