Bacterial death from treatment with fluoroquinolones and other lethal stressors

Karl Drlica et al.

Summary: Quinolones trap topoisomerases on DNA to block replication and growth, with DNA ends released at higher drug concentrations leading to chromosome fragmentation. Repair processes stimulated by DNA ends are energetically expensive, with ROS accumulation causing secondary damage even after quinolone removal. Interference with ROS accumulation can confer tolerance to various lethal agents.

Heterogeneous Host-Pathogen Encounters Coordinate Antibiotic Resilience in Mycobacterium tuberculosis

Richa Mishra et al.

Summary: The successful treatment of tuberculosis relies on eradicating Mycobacterium tuberculosis in the host, but the emergence of drug-resistant strains hampers antibiotic effectiveness. Host immunity creates diverse microenvironments exploited by the bacteria, amplifying pre-existing heterogeneity and impacting disease pathology and therapy outcomes. Understanding phenotypic heterogeneity may lead to new therapeutic strategies targeting host-pathogen interactions for TB cure.

Mycobacterium tuberculosis SufR responds to nitric oxide via its 4Fe-4S cluster and regulates Fe-S cluster biogenesis for persistence in mice

Kushi Anand et al.

Summary: The study reveals that the transcription factor SufR senses NO and promotes the persistence of Mycobacterium tuberculosis by mobilizing the Fe-S cluster biogenesis system. NO directly damages Fe-S clusters, resulting in the inability of Mtb to recover from a non-growing state, ultimately leading to a persistence defect in immune-activated host cells and murine lungs.

Four-Month Rifapentine Regimens with or without Moxifloxacin for Tuberculosis

S. E. Dorman et al.

Summary: The study found that a 4-month rifapentine-based regimen containing moxifloxacin was noninferior to the standard 6-month regimen in the treatment of tuberculosis, supporting the noninferiority hypothesis. This regimen was effective in both the microbiologically eligible population and the assessable population.

The Multifaceted Bacterial Cysteine Desulfurases: From Metabolism to Pathogenesis

Mayashree Das et al.

Summary: Living cells have developed a relay system to efficiently transfer sulfur from cysteine to various thio-cofactors, with multiple protein components regulating the flux of sulfur to prevent toxic accumulation. The first enzyme in this system, cysteine desulfurase (CSD), catalyzes the release of sulfane S from cysteine and transfers it to acceptor proteins for incorporation into thio-cofactors. CSD plays a role in bacterial core metabolic processes and is a potential target for therapy in human pathogens.
