Loss of heterochromatin and retrotransposon silencing as determinants in oocyte aging

Summary: Mammalian oocyte quality decreases with age due to the loss of heterochromatin histone marks, leading to impaired oocyte maturation. This loss also occurs in human oocytes. The age-related heterochromatin loss is associated with increased RNA processing, retrotransposon expression, DNA damage, and DNA repair proteins nuclear localization. Inhibiting the heterochromatin machinery in young oocytes causes maturation defects and elevated retrotransposon expression, while inhibiting retrotransposon reverse-transcriptase in older oocytes partially rescues their maturation defects. Activating the heterochromatin machinery in older oocytes upregulates constitutive heterochromatin, downregulates retrotransposon expression, and promotes maturation rates.

Summary: Failure of homologous chromosomes to recombine is a significant cause of human meiotic nondisjunction, particularly in maternal origin trisomies. Studies have found high levels of recombination failure in fetal ovarian samples, with a preponderance of MLH1-less homologs involving chromosomes 21 or 22. The presence of exchangeless chromosomes in cells is associated with a reduction in the total number of crossovers, indicating individuals with lower rates of meiotic recombination may be at increased risk of producing aneuploid offspring.

Summary: In this study, it was found that ovarian stimulation did not increase the levels of aneuploidy in blastocysts generated from OPU-IVP cycles in cattle. Most abnormalities consisted of meiotic trisomies. Blastocysts derived from in vivo donors were all euploid, indicating that maturation is primarily responsible for aneuploidy induction. Moreover, there was a high degree of concordance in the incidence of aneuploidy between different lineages of blastocysts.

Summary: Female fertility decreases with maternal age due to a decrease in oocyte quality and developmental competence. This study found that the transcriptome is more affected by age in in vitro matured (IVM-MII) oocytes compared to germinal vesicle stage (GV) oocytes. Specifically, genes related to chromosome segregation and RNA splicing showed increased representation with age in IVM-MII oocytes, while genes associated with mitochondrial activity displayed lower representation.

Generation of ovarian follicles from mouse pluripotent stem cells

Summary: Novel methods have been developed to generate functional mouse oocytes from pluripotent stem cell-derived primordial germ cell-like cells (PGCLCs) and fetal ovarian somatic cell-like cells (FOSLCs). When FOSLCs are aggregated with PGCLCs, they enter meiosis and produce oocytes capable of fertilization and development into live offspring, providing a method for in vitro oocyte production and follicle generation for a better understanding of mammalian reproduction.
