Proteomic Tools for the Quantitative Analysis of Artificial Peptide Libraries: Detection and Characterization of Target-Amplified PD-1 Inhibitors

Marina Gay et al.

Summary: We report a quantitative proteomics data analysis pipeline coupled with protein-directed dynamic combinatorial chemistry experiments, which enables rapid discovery and characterization of protein-protein interaction modulators. This study provides proof-of-concept for using this strategy in high-throughput search of improved drug-like peptide binders that block therapeutically relevant protein-protein interactions.

Fuel-Driven Dynamic Combinatorial Libraries

Christine M. E. Kriebisch et al.

Summary: Dynamic combinatorial libraries involve molecules reacting reversibly to form intricate networks under thermodynamic or kinetic control. The dynamics of the library are dominated by transacylation, an equilibrium reaction, while fuel-driven activation dictates the library's dynamics, a nonequilibrium reaction. Self-assembly can play a significant role in affecting reaction kinetics through feedback mechanisms.

Hit-optimization using target-directed dynamic combinatorial chemistry: development of inhibitors of the anti-infective target 1-deoxy-d-xylulose-5-phosphate synthase

Ravindra P. Jumde et al.

Summary: Target-directed dynamic combinatorial chemistry (tdDCC) was used to identify and optimize ligands for the anti-infective target DXPS, resulting in acylhydrazone-based inhibitors. The tailored tdDCC runs provided insights into the structure-activity relationship of these novel inhibitors, with competition runs offering important information about their mode of inhibition. This approach has the potential to expedite drug-discovery and may be applicable to various biological targets.
