Advancing the Logic of Chemical Synthesis: C-H Activation as Strategic and Tactical Disconnections for C-C Bond Construction

Summary: The design of synthetic routes by retrosynthetic logic is influenced by available transformations. Transition-metal-catalyzed C-H activation is a powerful strategy for C-C bond formation, but its uptake in total synthesis has been tepid due to synthetic intractability and a lack of comprehensive guidelines. This Review addresses these issues and offers a guide to identify retrosynthetic opportunities to generate C-C bonds by C-H activation processes. Comparing traditional approaches and recent C-H activation methods, this Review demonstrates more efficient retrosynthetic strategies enabled by C-H activation for C-C bond construction.

Summary: The cobalt-catalyzed Suzuki biaryl cross-coupling reaction using alkoxide bases, with strict stoichiometric control and selection of the right boron ester, is shown to be achievable. Excessive alkoxide bases or lithium additives can inhibit or poison the catalyst, while a neopentane diol-based boron ester is crucial for optimal performance.Mechanistic and computational studies have been carried out to investigate the reaction mechanism and explain unexpected observations.

Beyond hydrogen bonding: recent trends of outer sphere interactions in transition metal catalysis

Summary: Homogeneous catalytic reactions are typically controlled by the stereoelectronic nature of ligands binding to metals. The first coordination sphere effects and second coordination sphere effects, inspired by enzymes, rely on interactions such as hydrogen bonding for fine control of activity and selectivity. These interactions are being rationally implemented to control reactivity in a remote fashion, similar to enzymes, for challenges that cannot be purely solved by hydrogen bonding.
