Silencing LDHC in breast cancer cell lines leads to cellular abnormalities, DNA damage, and increased apoptosis, as well as microtubule destabilization leading to mitotic catastrophe. LDHC silencing also enhances sensitivity to drugs causing DNA damage or inhibiting repair mechanisms.

Summary: Analysis of the testis proteome in Ubb knockout mice revealed 564 proteins significantly differentially expressed, with 36 downregulated proteins involved in various stages of spermatogenesis. Functional gene ontology analysis identified decreased levels of piRNA metabolic process-related proteins, such as Piwil2 and Tdrd1, in Ubb-null testes. Protein-protein interaction mapping showed that 24 testis development-related proteins, including Hsp90aa1, Eef1a1, and Pabpc1, were directly impacted by the depletion of ubiquitin, with mRNA levels of these proteins reduced in Ubb-knockout testes resembling the global downregulation of piRNA-metabolic gene expression at transcriptional and post-transcriptional levels.
