Simultaneous Counting of Molecules in the Halo and Dense-Core of Nanovesicles by Regulating Dynamics of Vesicle Opening

Xiulan He et al.

Summary: In the presence of chaotropic anions, the opening of nanometer biological vesicles becomes a two-step process, allowing for independent counting of molecules in each subvesicular compartment. The study revealed a 68% catecholamine binding to the dense-core and differentiated two populations of large dense-core vesicles, with differences in biogenesis speculated to be caused by an increase in electrostatic attraction.

New trends in enzyme-free electrochemical sensing of ROS/RNS. Application to live cell analysis

Daniel Rojas et al.

Summary: The ubiquity and importance of ROS and RNS in cellular signaling, disease development, and death have led to a significant interest in their detection and quantification. Electrochemical sensors are particularly suitable for the detection of ROS/RNS due to their high sensitivity and miniaturization, allowing for in situ and real-time detection. Nanomaterial-based enzyme-free electrochemical sensors have superior sensitivity, selectivity, stability, and catalytic activities. Moreover, their integration into nanoelectrodes, lab-on-chips, microfluidic systems, and stretchable electrodes enables the determination of ROS/RNS in individual cells, cell organelles, or cell populations under different experimental conditions that are difficult to achieve using classical detection methods.

Engineered Aptamer-Organic Amphiphile Self-Assemblies for Biomedical Applications: Progress and Challenges

Hongjie Xiong et al.

Summary: Nucleic acid aptamers are used as targeting ligands in biomedical field due to their specific molecular recognition and low immunogenicity. Aptamer-organic amphiphile self-assemblies (AOASs) have enhanced target-binding ability and resistance to nuclease, making them powerful platforms for bioapplications such as drug delivery and cancer therapy. Manipulating the hydrophilicity/hydrophobicity ratio of AOASs can lead to smarter biomedical functions.

Modulation of mitochondrial electron transport chain by pyroptosis for tumor destruction

Penghao Ji et al.

Summary: Programmed cell death (PCD) plays a crucial role in combating tumors in the field of biomedicine. However, malignant tumor cells often develop resistance to commonly used anticancer drugs. A newly designed nanoassembly OA@IR820 targets mitochondria and releases ATP synthase inhibitor OA, leading to ATP synthesis inhibition and mitochondrial dysfunction, thereby promoting oxidative stress-mediated pyroptosis and offering a potential therapeutic strategy for malignant tumor treatment.

Yingwen Wang et al.

Summary: In this study, a novel impedimetric aptasensor was developed for the discriminative detection of intracellular and extracellular ATP, achieving improved sensitivity and selectivity through the use of antimicrobial peptide-functionalized magnetic particles. By introducing magnetically controlled M-AMPs into the detection system, mild and rapid bacterial disintegration was achieved for effective ATP detection. The proposed aptasensor exhibited excellent performance with a wide linear range and low detection limit, providing a convenient and label-free approach for discriminative intracellular and extracellular ATP detection in various bacterial samples and E. coli biofilms.
