Global burden of bacterial antimicrobial resistance in 2019: a systematic analysis

Christopher J. L. Murray et al.

Summary: Antimicrobial resistance (AMR) poses a significant threat to global human health, and this study provides the most comprehensive estimates of AMR burden to date. By estimating deaths and disability-adjusted life-years (DALYs) associated with bacterial AMR in 204 countries and territories in 2019, the study highlights the impact of resistance and the leading pathogen-drug combinations contributing to it.

MicroRNA sequence codes for small extracellular vesicle release and cellular retention

Ruben Garcia-Martin et al.

Summary: Research has shown that miRNAs have sorting sequences that determine their secretion in sEVs, different cell types preferentially use specific sorting sequences to define their sEV miRNA profile. Insertion or deletion of these sequences affects miRNA secretion. RNA-binding proteins Alyref and Fus are involved in the mechanism, leading to enhanced miRNA delivery and inhibition of target genes in distant cells.

Widespread of Potential Pathogen-Derived Extracellular Vesicles Carrying Antibiotic Resistance Genes in Indoor Dust

Yifei Qin et al.

Summary: This study investigated the presence of ARGs and MGEs in EVs from indoor dust in restaurants, kindergarten, dormitories, and vehicles. The results showed that EVs contained a significant amount of ARGs and MGEs, including resistance genes to multiple classes of antibiotics. Some ARGs were found to co-occur with MGEs. The taxonomic composition of EVs was different from their original dust microbiota, and a portion of the EV-associated DNA was predicted to come from potential pathogens.

Systemically circulating bacterial extracellular vesicles: origin, fate, and function

An Hendrix et al.

Summary: Bacteria contribute to human physiology and pathology through the production of membrane-enclosed biomolecules known as bacterial extracellular vesicles (BEVs). Recent research has shown that BEVs, as functional products of bacteria, enter the systemic circulation. This article highlights the current understanding of the origin, translocation, distribution, function, and elimination of systemically circulating BEVs, and identifies areas where further research is needed to uncover the role of BEVs in health and disease.

Single Particle Automated Raman Trapping Analysis of Breast Cancer Cell-Derived Extracellular Vesicles as Cancer Biomarkers

Jelle Penders et al.

Summary: The study focuses on extracellular vesicles (EVs) and introduces a high-throughput label-free EV analysis method for cancer diagnosis and monitoring, eliminating interpreter bias. By developing a dedicated device and using dimensional reduction analysis (DRA), a convenient and comprehensive approach for comparing multiple EV spectra is demonstrated. The study shows the high sensitivity and specificity of the dedicated SPARTA system in differentiating cancer and noncancer EVs, and its consistent predictive ability across multiple EV isolations.

Dinoroseobacter shibae Outer Membrane Vesicles Are Enriched for the Chromosome Dimer Resolution Site dif

Hui Wang et al.

Summary: Outer membrane vesicles (OMVs) secreted by Dinoroseobacter shibae during cell division contain DNA enriched for specific gene regions, potentially repaired from overreplication. The vesicle proteome includes proteins interacting with peptidoglycan during division. The FtsK-dif-XerC/XerD molecular machinery may provide a novel route for DNA incorporation into OMVs.

DEHP-elicited small extracellular vesicles miR-26a-5p promoted metastasis in nearby normal A549 cells

Yifei Qin et al.

Summary: DEHP exposure promotes the secretion of miR-26a-5p in sEV, subsequently enhancing the EMT, migration, and invasion ability in neighboring normal cells.

Temperature Influences the Composition and Cytotoxicity of Extracellular Vesicles in Staphylococcus aureus

Paul Briaud et al.

Summary: This study demonstrates that temperature variations during Staphylococcus aureus infection process can significantly impact the production, composition, and function of extracellular vesicles (EVs). Higher temperatures promote the packaging of virulence factors in EVs, while lower temperatures lead to increased toxicity towards macrophages. The findings highlight the importance of temperature-dependent interactions between EVs and host cells during S. aureus transition from colonization to virulence.

Archaeal extracellular vesicles are produced in an ESCRT-dependent manner and promote gene transfer and nutrient cycling in extreme environments

Junfeng Liu et al.

Summary: Membrane-bound extracellular vesicles (EVs) are secreted by cells from all three domains of life, carrying a diverse proteome and DNA, and can support bacterial growth in extreme environments. The production of EVs in bacteria is mediated by the ESCRT machinery, with a prime role of ESCRT-III-1 and ESCRT-III-2 proteins in EV budding. EVs have deep evolutionary roots and play a crucial role in horizontal gene transfer and nutrient cycling in extreme environments.

Visualizing transfer of microbial biomolecules by outer membrane vesicles in microbe-host-communication in vivo

Miriam Bittel et al.

Summary: Our study shows that bacterial OMVs can serve as a biological shuttle system for horizontal transfer of functional biomolecules between bacteria and mammalian host cells. Introducing E. coli engineered to express Cre-recombinase into mice demonstrated the transfer of bacterial OMVs to recipient cells in the gut and beyond. This evidence provides a method and proof of principle for inter-kingdom communication facilitated by OMVs.
