Dual-Aptamer-Targeted Immunomagnetic Nanoparticles to Accurately Explore the Correlations between Circulating Tumor Cells and Gastric Cancer

Chenglin Li et al.

Summary: It has been acknowledged that circulating tumor cells (CTCs) serve as promising biomarkers for cancer diagnosis and prognosis. This study proposes a simple and cost-effective method using dual-aptamer-modified immunomagnetic particles to efficiently capture heterogeneous CTCs in gastric cancer patients. The results show that this method can accurately recognize CTCs with both high and low expression of epithelial cell adhesion molecule (EpCAM) and protein tyrosine kinase 7 (PTK7), and the detected CTC numbers are closely related to chemotherapy sensitivity, diagnosis, stage, and distant metastasis in gastric cancer patients.

Summary: Colorectal cancer is a major global public health challenge with higher incidence rates in developed countries and increasing trends in middle- and low-income countries. The rise of early-onset colorectal cancer is also seen. Increasing awareness, promoting healthy lifestyle choices, and implementing global screening programs are crucial in reducing the morbidity and mortality of colorectal cancer in the future.

Summary: Circulating tumor cells (CTCs) are shed from tumors into the bloodstream and have the potential to provide important insights into cancer detection and monitoring. CellSearch is the only FDA-approved technology for determining the prognosis of patients with advanced breast, prostate, and colorectal cancers, but new label-free enrichment technologies are emerging.

Characterisation of circulating tumour cell phenotypes identifies a partial-EMT sub-population for clinical stratification of pancreatic cancer

Alexander Semaan et al.

Summary: DEP-FFF was used to isolate CTCs in PDAC patients, revealing four distinct sub-populations including pEMT-CTCs, which were correlated with disease progression, survival, and recurrence. This highlights the importance of immunophenotyping and quantifying specific CTC sub-populations in PDAC.

Summary: The study demonstrated the potential clinical value of isolating and characterizing heterogeneous sub-populations of CTCs in PDAC patients. Both EMT-related CTCs and hENT-1 expression in CTCs were identified as potential prognostic biomarkers for recurrence free survival in resected patients receiving adjuvant gemcitabine-based chemotherapy.

Circulating tumor cells as a promising target for individualized drug susceptibility tests in cancer therapy

Daniel J. Smit et al.

Summary: CTCs play a crucial role in cancer metastasis and their analysis in liquid biopsies can be valuable for prognosis prediction and real-time therapy monitoring. Research on CTCs may lead to new treatment strategies for preventing metastasis or maintaining stable diseases. Studies on inhibitors targeting CTCs have shown promising results, suggesting CTCs may be useful for personalized drug susceptibility testing.

Functional Implications of the Dynamic Regulation of EpCAM during Epithelial-to-Mesenchymal Transition

Taylor C. Brown et al.

Summary: Epithelial cell adhesion molecule (EpCAM) is a transmembrane glycoprotein that plays a crucial role in epithelial tissue homeostasis and is a target of molecular therapies for tumor biology. Its expression is frequently high in epithelial cancers and silenced in mesenchymal cancers. EpCAM's functional impact on tumor biology is multifunctional and depends on the predominant oncogenic signaling pathways in different cancer types. Its regulation during epithelial-to-mesenchymal transition (EMT) involves downregulation by extracellular signal-regulated kinases (ERK) and EMT transcription factors, as well as regulated intramembrane proteolysis (RIP).

Circulating tumor cells: biology and clinical significance

Danfeng Lin et al.

Summary: CTCs, tumor cells that have sloughed off the primary tumor and circulate in the blood, have potential for identifying targets against cancer metastasis. Detecting these rare CTCs in the massive pool of blood cells is challenging, but emerging detection technologies have deepened our understanding of their biology and clinical applications. Molecular markers, such as epithelial cell adhesion molecule, play a key role in CTC detection, with properties like epithelial-to-mesenchymal transition and stemness indicating increased metastatic capacity.
