Differential reprogramming of breast cancer subtypes in 3D cultures and implications for sensitivity to targeted therapy

Three-Dimensional Aggregated Spheroid Model of Hepatocellular Carcinoma Using a 96-Pillar/Well Plate

Summary: The ASM creates large single spheroids by aggregating cells in Matrigel attached to the surface of 96-pillar plates, addressing the lack of cell-to-cell interaction issue in traditional 3D cell cultures. In drug screening assays, the ASM demonstrated higher drug resistance and greater expression of cancer markers associated with proliferation, tight junction formation, and epithelial cell identity compared to CSM and 2D cell culture model. The ASM also exhibited limited drug penetration in tissues in vivo, providing more instructive data for in vitro drug screening assays of tumor cells and improved prediction of efficacious drugs in HCC patients.
