T-cell receptor sequencing specifies psoriasis as a systemic and atopic dermatitis as a skin-focused, allergen-driven disease

Lennart M. Roesner et al.

Summary: This study compares T cells from lesional skin and blood of patients with atopic dermatitis (AD) and psoriasis, and finds that analyzing the T-cell receptor (TCR) sequences that drive skin-homing T cells helps to clarify the extent to which these infiltrating T cells are antigen-specific.

Mild atopic dermatitis lacks systemic inflammation and shows reduced nonlesional skin abnormalities

Helen He et al.

Summary: This study found that markers of epidermal hyperplasia and T-cell/dendritic cell infiltration were increased in the skin tissues of all AD patients, regardless of disease severity. Levels of T(H)2/T(H)22 cytokines were significantly elevated in both lesional and nonlesional skin of all patients, while T(H)1 and T(H)17 markers were increased only in lesional skin of mild AD patients. Blood profiles showed gradual increases in inflammatory markers in moderate and severe AD patients, but no significant differences were observed in mild AD patients compared to healthy controls.

Single-cell RNA sequencing of psoriatic skin identifies pathogenic Tc17 cell subsets and reveals distinctions between CD8+ T cells in autoimmunity and cancer

Jared Liu et al.

Summary: This study utilized single-cell RNA sequencing to compare the transcriptomic heterogeneity of CD8(+) T cells in psoriatic and healthy skin, identifying several inflammatory subsets enriched in psoriatic skin and two Tc17 cell subsets associated with disease severity. Despite high expression of coinhibitory receptors, the psoriatic Tc17 cells exhibited upregulated cytokine, cytolytic, and metabolic activity, distinguishing them from an exhaustion program observed in melanoma-infiltrating CD8(+) T cells.

Developmental cell programs are co-opted in inflammatory skin disease

Gary Reynolds et al.

Summary: The skin establishes a complex cellular network early in embryonic development to provide biophysical and immunological protection. The study found an enrichment of innate immune cells in skin during the first trimester, as well as clonal expansion of disease-associated lymphocytes in atopic dermatitis and psoriasis. Additionally, a reemergence of prenatal vascular endothelial cell and macrophage cellular programs was observed in atopic dermatitis and psoriasis lesional skin.

Persistence of mature dendritic cells, TH2A, and Tc2 cells characterize clinically resolved atopic dermatitis under IL-4Rα blockade

Christine Bangert et al.

Summary: Therapeutic options for autoimmune diseases typically involve immunosuppressive agents, but sustained clinical benefit is rare. A study on atopic dermatitis patients treated with dupilumab found decreases in skin immune cell counts and normalization of transcriptomic dysregulation. However, specific immune cell populations persisted after clinical remission, indicating a persisting disease memory.

Single-cell transcriptomics applied to emigrating cells from psoriasis elucidate pathogenic versus regulatory immune cell subsets

Jaehwan Kim et al.

Summary: This study utilized single-cell transcriptomics to analyze emigrating cells from the skin, revealing gene expression profiles of pathogenic and regulatory immune cell subsets in individuals with psoriasis. The findings also highlighted differences in key signaling pathways in psoriasis.

Does the epithelial barrier hypothesis explain the increase in allergy, autoimmunity and other chronic conditions?

Cezmi A. Akdis

Summary: The increase in allergic, autoimmune, and chronic diseases may be related to epithelial barrier-damaging agents associated with industrialization, urbanization, and modern life. These diseases are more prevalent in industrialized countries and are on the rise in developing countries, affecting over one billion people globally.
