Bioprinted Constructs that Mimic the Ossification Center Microenvironment for Targeted Innervation in Bone Regeneration

Wentao Li et al.

Summary: The NGF@Lap constructs, created through bioprinting technology, simulate the ossification center microenvironment and promote innervation, ultimately leading to tissue regeneration. They enhance CGRP expression and secretion, regulate osteogenesis and inhibit adipogenesis through a specific pathway, and in vivo, they increase neurovascular network density and promote osteogenic differentiation of bone marrow mesenchymal stem cells.

Extracellular vesicles derived from neural EGFL-Like 1-modified mesenchymal stem cells improve acellular bone regeneration via the miR-25-5p-SMAD2 signaling axis

Yanhua Lan et al.

Summary: This study reveals that extracellular vesicles (EVs) derived from Nell1-modified bone marrow stem cells (BMSCs) have a stronger ability to promote BMSC osteogenesis due to the downregulation of miR-25-5p. Additionally, the 3D-Nell1/EV hydrogel system is shown to be beneficial for bone regeneration in vivo, possibly because of the slow and continuous release of EVs at a high concentration in the bone defect area.

The effects of microgravity on bone structure and function

Joey Man et al.

Summary: This review discusses the issue of bone loss in humans and animals exposed to microgravity in space. While space experiments are limited, ground research that simulates microgravity offers insights into the molecular mechanisms of bone loss and the development of more effective preventative and therapeutic measures. However, there are still knowledge gaps in understanding the exact mechanisms of microgravity-induced bone loss.

Summary: This study discusses the regulatory mechanisms of ncRNAs in bone defect repair and reviews the current RNA delivery vectors and scaffolds. Non-coding RNAs play important roles in regulating gene expression and delivering them to the site of bone loss could be a potential therapeutic option. However, there are still challenges and potential solutions for future clinical applications.

Stratified-structural hydrogel incorporated with magnesium-ion-modified black phosphorus nanosheets for promoting neuro-vascularized bone regeneration

Yan Xu et al.

Summary: Angiogenesis and neurogenesis play important roles in bone repair. This study focuses on the development of a bilayer hydrogel that mimics the periosteum and incorporates magnesium-ion-modified black phosphorus nanosheets to enhance neurovascularized bone regeneration. The results demonstrate that this bilayer hydrogel platform can significantly promote angiogenesis and neurogenesis, as well as enhance the activity and osteogenic differentiation of bone marrow mesenchymal stem cells. Furthermore, in vivo experiments show that the bilayer hydrogel scaffolds effectively enhance early vascularization and neurogenesis, leading to successful bone regeneration and remodeling.

The dual-effects of PLGA@MT electrospun nanofiber coatings on promoting osteogenesis at the titanium-bone interface under diabetic conditions

Zijie Wang et al.

Summary: This study investigates the effects of melatonin on the titanium-bone interface in diabetic conditions. The results show that the nanofiber coating created using PLGA@MT-Ti promotes osteogenesis and osteointegration by activating the BMP-4/WNT pathway and attenuating ROS overproduction. This provides a novel strategy for biomaterial modification and biofunctionalization under diabetic conditions.

Crosstalk between Bone and Nerves within Bone

Qian-Qian Wan et al.

Summary: This review comprehensively analyzes the possible crosstalk between bone and intrabony peripheral nerves, highlighting the role of peripheral nerves in bone development and repair through signals such as neurotransmitters, neuropeptides, and neurotrophins. In return, bone supports nerves mechanically and offers a protective environment. This crosstalk is not limited to physiological conditions, but also plays a role in various bone diseases, providing potential avenues for tissue engineering and disease treatment.
