Engineered HaloTag variants for fluorescence lifetime multiplexing

Michelle S. Frei et al.

Summary: HaloTag variants offer unique brightness and fluorescence lifetimes compared to HaloTag7 when labeled with rhodamines, enabling multiplexed imaging and creating cell cycle indicators. These variants are powerful tools for fluorescence microscopy and can be used to generate fluorescence lifetime-based biosensors, with the brightest variant showing up to 40% higher brightness in live-cell imaging applications.

Transient Fluorescence Labeling: Low Affinity-High Benefits

Maxim M. Perfilov et al.

Summary: Fluorescent labeling is a well-established method for visualizing cellular structures and dynamics, with super-resolution microscopy bypassing the diffraction limit in image resolution. Transiently interacting labels can provide high photostability through constant supply, while also interfering less with the intrinsic dynamics and cellular functions of labeled molecules.

Engineering of a fluorescent chemogenetic reporter with tunable color for advanced live-cell imaging

Hela Benaissa et al.

Summary: Fluorescent reporters covering the visible spectrum are essential for imaging living cells and organisms. The authors have developed a collection of fluorogenic chromophores spanning from blue to red using a single engineered protein tag. This tool allows for tuning fluorescence color and properties, providing versatility for imaging proteins in live cells and optimizing FRET biosensors.

MINSTED fluorescence localization and nanoscopy

Michael Weber et al.

Summary: MINSTED is a fluorescence localization and super-resolution microscopy concept based on STED, offering high spatial precision and resolution while reducing fluorophore bleaching. By conducting 200-1,000 detections per fluorophore, a localization precision of 1-3 nm can be achieved, resulting in a significant improvement in far-field microscopy resolution over the diffraction limit.

Kinetic and Structural Characterization of the Self-Labeling Protein Tags HaloTag7, SNAP-tag, and CLIP-tag

Jonas Wilhelm et al.

Summary: The study compared the labeling kinetics and substrate specificities of self-labeling protein tags (SLPs) HaloTag7, SNAP-tag, and CLIP-tag. It was found that HaloTag7 reaches almost diffusion-limited labeling rate constants with some substrates, while SNAP-tag labeling rate constants are less affected by the structure of the label. Crystal structures of HaloTag7 and SNAP-tag labeled with fluorescent substrates helped explain their substrate preferences and aided in the design of substrates with improved labeling kinetics.

Triple-Color STED Nanoscopy: Sampling Absorption Spectra Differences for Efficient Linear Species Unmixing

Mariano Gonzalez Pisfil et al.

Summary: STED technique allows visualization of biological structures within cells below diffraction limit by exploiting spectral differences in absorption of fluorescent labels for simultaneous detection of multiple species with close absorption maxima. This novel approach using multiple excitation wavelengths in nanosecond pulsed interleaved excitation mode decomposes acquired images into single species contributions through a linear unmixing algorithm. The introduced image correlation map (ICM) facilitates identification of single species regions for generating necessary single species reference patterns, showing high speed species separation quality for up to three species in cellular STED imaging.

Long-term STED imaging of membrane packing and dynamics by exchangeable polarity-sensitive dyes

Pablo Carravilla et al.

Summary: Understanding the nanoscale organization and dynamics of plasma membrane in living cells requires microscopy techniques with high spatial and temporal resolution. The polarity-sensitive exchangeable plasma membrane probe NR4A based on Nile red allows real-time super-resolved quantification of membrane biophysical parameters with high temporal and spatial resolution, as well as long acquisition times.
