Large-scale deep multi-layer analysis of Alzheimer's disease brain reveals strong proteomic disease-related changes not observed at the RNA level

Erik C. B. Johnson et al.

Summary: This study analyzed the proteomes of over 1,000 brain tissues and identified new protein co-expression modules associated with Alzheimer's disease. These modules were highly conserved across cohorts and brain regions and revealed a proteopathic nature of the disease. The study also found disease-associated modules unique to the proteome, which could serve as potential therapeutic targets and biomarkers for Alzheimer's disease.

Acutely Inhibiting AQP4 With TGN-020 Improves Functional Outcome by Attenuating Edema and Peri-Infarct Astrogliosis After Cerebral Ischemia

Chengfeng Sun et al.

Summary: Acute inhibition of AQP4 using TGN-020 can alleviate brain edema, promote neurological recovery, and attenuate peri-infarct astrocyte reactivity and AQP4 depolarization. In addition, ADCuh can reflect AQP4 polarization.

Activated endothelial cells induce a distinct type of astrocytic reactivity

Xavier Taylor et al.

Summary: Injured endothelial cells can induce neurotoxic astrocytes characterized by genetic signatures associated with extracellular matrix remodeling. These astrocytes, activated by endothelial cells, are distinct from those activated by other cell types and are associated with vascular amyloid deposits.

Rapid multilayer microfabrication for modeling organotropic metastasis in breast cancer

Tae Joon Kwak et al.

Summary: Understanding the interaction between TNBC and blood vasculatures is crucial in studying and treating TNBC metastasis, particularly in the process of TNBC extravasation from blood vessels to determine the final site of metastasis. Modeling studies have shown that bone and lungs are common sites of breast cancer metastasis, and organ-specific parenchymal cells play a key role in the selective extravasation of TNBC cells in bone and lung microenvironments.

Stagnation of glymphatic interstitial fluid flow and delay in waste clearance in the SOD1-G93A mouse model of ALS

Mikako Hirose et al.

Summary: The study indicates that abnormal expression and mislocalization of AQP4 in SOD1(G93A) mice result in dysfunction of the glymphatic system, leading to delayed waste clearance and exacerbation of ALS disease progression.

Acute systemic LPS-exposure impairs perivascular CSF distribution in mice

Oscar Manouchehrian et al.

Summary: The study aimed to measure the effect of acute inflammation on the glymphatic system by analyzing the distribution of CSF tracers after LPS injections. The results showed a significant decrease in perivascular CSF tracer flow and penetration into the parenchyma following LPS injections, without impacting AQP4 polarization, cytokines, astroglial and BBB markers, cerebral blood flow, or respiration.

The Neuro-Glial-Vascular Unit: The Role of Glia in Neurovascular Unit Formation and Dysfunction

Elisabeth C. Kugler et al.

Summary: The neurovascular unit is a complex multi-cellular structure consisting of various cell types such as endothelial cells, neurons, glia, smooth muscle cells, and pericytes. While neurovascular coupling is a key function of the NVU, the unit's functionality extends beyond this coupling to include signaling, metabolism, and homeostasis across different components. Glial cells have gained increased attention within the NVU and play central roles in its functionality and disease pathogenesis.

The Glymphatic System (En)during Inflammation

Frida Lind-Holm Mogensen et al.

Summary: The glymphatic system is a fluid-transport system in the brain that plays a role in neurological diseases and neuroinflammation. It facilitates waste clearance in the brain, requires fluid exchange between perivascular spaces and the neuropil, and transports fluid back to the vasculature through lymphatic vessels.
