HDL cholesterol concentrations and risk of atherosclerotic cardiovascular disease - Insights from randomized clinical trials and human genetics

Emilie Westerlin Kjeldsen et al.

Summary: Over the past seven decades, studies have observed an inverse association between HDL cholesterol concentrations and atherosclerotic cardiovascular disease (ASCVD) risk. However, recent evidence from human genetics and randomized clinical trials suggest that targeting HDL cholesterol concentrations may not be a viable approach for preventing ASCVD.

MEF2A Is the Trigger of Resveratrol Exerting Protection on Vascular Endothelial Cell

Benrong Liu et al.

Summary: Resveratrol enhances the expression of MEF2A and the upregulation of MEF2A is required for the protective effects of resveratrol on vascular endothelial cells. Silencing MEF2A can impair the protective function of resveratrol and aggravate atherosclerotic lesion.

Coronary Artery Disease Genetics Enlightened by Genome-Wide Association Studies

Thorsten Kessler et al.

Summary: Many cardiovascular diseases have a strong genetic component, with large-scale genetic studies identifying genomic loci that influence the risk of specific diseases. While most candidate genes at these loci are not traditionally associated with disease risk factors, experimental research is uncovering novel and unexpected mechanisms. Genome-wide association studies have already led to the development of new treatments, particularly in lipid metabolism. Additionally, polygenic risk scores may have clinical utility in risk prediction and personalized treatment, especially in lipid metabolism.

Correlation between plasma homocysteine and first myocardial infarction in young patients: Case-control study in Constanta County, Romania

Cristina Nedelcu et al.

Summary: Elevated plasma homocysteine levels are associated with increased risk of atherosclerosis and coronary heart disease. The study conducted in Romania revealed a strong association between high homocysteine levels and first acute myocardial infarction in young patients, particularly in conjunction with smoking, chronic kidney disease (CKD), and to a lesser extent with diabetes mellitus (DM) and hypertension. Data analysis suggested that plasma homocysteine could be a potential risk factor for myocardial infarction in young individuals.

Association of CETP Gene Variants with Atherogenic Dyslipidemia Among Thai Patients Treated with Statin

Pornpen Srisawasdi et al.

Summary: Genetic variants in CETP, specifically rs3764261 and rs708272, are associated with dyslipidemia and may increase susceptibility to atherosclerotic cardiovascular disease. Testing for these variants could help tailor lipid management in statin-treated patients to reduce residual ASCVD risk.

Association of OLR1 gene polymorphisms with the risk of coronary artery disease: A systematic review and meta-analysis

Pouya Salehipour et al.

Summary: The study found a significant association between OLR1 polymorphisms in the haplotype block, especially rs1050283 and rs3736235, with the risk of CAD. It is suggested that precise determination of disease association with polymorphisms in a haplotype requires investigation of all SNPs rather than a single SNP in that specific haplotype.

Clinical utility of polygenic risk scores for coronary artery disease

Derek Klarin et al.

Summary: In recent years, significant progress has been made in the discovery of genes related to the risk of coronary artery disease, and polygenic risk scores are widely used for predicting disease occurrence and guiding clinical treatment. These scores are also used for screening high-risk patients, identifying treatment-beneficial populations, and designing effective clinical trial protocols.

Sialic acid as a potential biomarker for cardiovascular disease, diabetes and cancer

Summary: Elevated sialic acid concentration in plasma and serum is positively correlated with the presence of cardiovascular disease, diabetes, and malignant tumors. While not a specific marker, sialic acid shows promise as a marker for monitoring disease progression and treatment effectiveness.

Lipoprotein Lipase Gene Polymorphisms Are Associated with Myocardial Infarction Risk: A Meta-Analysis

Kunhui He et al.

Summary: The meta-analysis suggests that LPL HindIII and S447X polymorphisms may be protective factors for myocardial infarction, while PvuII is not associated. There are ethnic differences in the response to these polymorphisms, indicating the need for larger case-control studies to confirm the results.

Current Diagnosis and Management of Tangier Disease

Masahiro Koseki et al.

Summary: Tangier disease is a genetic disorder caused by dysfunctional mutation of the ABCA1 gene, leading to extremely low levels of HDL cholesterol. Common physical signs in patients include orange-colored pharyngeal tonsils and hepatosplenomegaly. The disease is associated with significantly reduced HDL-C levels and an increased risk of premature coronary artery disease.

Summary: Genomic sequencing advances have expanded our understanding of the genetic factors contributing to coronary artery disease, both monogenic and polygenic.
