Very long-chain saturated fatty acids and diabetes and cardiovascular disease

Rozenn N. Lemaitre et al.

Summary: This review summarizes the associations of very long-chain saturated fatty acids (VLSFAs) with cardiovascular disease outcomes and type 2 diabetes. It found that higher levels of circulating VLSFAs are associated with lower risks of heart failure, atrial fibrillation, coronary heart disease, mortality, sudden cardiac arrest, type 2 diabetes, and better aging. The associations of VLSFAs with cardiovascular diseases may be mediated by ceramides and sphingomyelins, while the associations with diabetes may be confounded by triglyceride and circulating palmitic acid.

Review Amino acid transporters as modulators of glucose homeostasis

Stefan Broer

Summary: Amino acids play a crucial role in modulating glucose homeostasis through regulating insulin release, protein synthesis, cell proliferation, and metabolism. Amino acid transporters are key regulators in different tissues and cells, controlling amino acid uptake, insulin release, and gluconeogenesis.

Investigating the Postprandial Metabolome after Challenge Tests to Assess Metabolic Flexibility and Dysregulations Associated with Cardiometabolic Diseases

Gaia Lepine et al.

Summary: This review discusses the use of metabolomics analyses in assessing phenotypic flexibility in response to different nutritional challenge tests. The findings reveal subtle metabolic dysregulations during the postprandial period, particularly in populations with increased cardiometabolic risk. The characterization of postprandial metabolism and phenotypic flexibility shows promise in understanding the onset of cardiometabolic diseases.

MOLECULAR PHYSIOLOGY OF BILE ACID SIGNALING IN HEALTH, DISEASE, AND AGING

Alessia Perino et al.

Summary: Bile acids have become important signaling molecules in the body, enabling inter-tissue communication and exerting pleiotropic physiological effects. The chemical variety of bile acids, largely determined by the gut microbiome, allows for complex adaptive responses in the body. This review discusses the mechanisms by which bile acid receptors coordinate various aspects of physiology and presents new therapeutic strategies for diseases related to pathological bile acid signaling.

Circulating cell adhesion molecules in metabolically healthy obesity

Arij Mulhem et al.

Summary: The study found that individuals with metabolically healthy obesity (MHO) have higher circulating cell adhesion molecules (CAMs) compared to metabolically healthy lean (MHL) individuals. While insulin sensitive (IS) MHO have lower CAM concentrations than insulin resistant (IR) MHO, they are still higher than MHL. Circulating adhesion molecules were significantly higher in individuals with metabolically unhealthy obesity (MUO) compared to other groups.

Regulation of Postabsorptive and Postprandial Glucose Metabolism by Insulin-Dependent and Insulin-Independent Mechanisms: An Integrative Approach

George D. Dimitriadis et al.

Summary: Glucose homeostasis is regulated by insulin, with key roles played by the liver, kidneys, skeletal muscle, and adipose tissue. Insulin resistance during sleep helps maintain normal blood glucose levels by switching to fatty acid oxidation in muscles and modulating glucose production from the liver and kidneys. Postprandial glucose fluctuations are optimally regulated by a combination of various mechanisms, including the liver's critical role in disposing ingested glucose and preventing acute increases in blood glucose levels after meals.

Insulin action, type 2 diabetes, and branched-chain amino acids: A two-way street

Phillip J. White et al.

Summary: The association of obesity and insulin resistance with increased levels of branched-chain and aromatic amino acids and decreased glycine levels is well-established. Human metabolomics and genetic studies have further confirmed these observations, with preclinical studies identifying mechanisms involved in amino acid homeostasis disruption and its connection to dysregulated glucose and lipid metabolism, particularly in the development of insulin resistance, type 2 diabetes, and cardiovascular diseases.

Advantages of Studying the Metabolome in Response to Mixed-Macronutrient Challenges and Suggestions for Future Research Designs

Jennifer L. LaBarre et al.

Summary: Evaluating the postprandial response to a dietary challenge containing all macronutrients may offer stronger insights into metabolic health than a fasted measurement. Metabolomic profiling deepens the understanding of the homeostatic and adaptive response to a dietary challenge. Studies assessing the postprandial response of the metabolome highlight the importance of individual amino acids and fatty acids in the metabolic process.

Plasma Bile Acid Profile in Patients with and without Type 2 Diabetes

Alessandro Mantovani et al.

Summary: This study reveals marked differences in plasma bile acid profiles between patients with and without T2DM, with higher levels of both primary and secondary bile acids in T2DM patients, as well as specific bile acid species showing significant associations with T2DM status. Further research is needed to elucidate the potential interplay between these differences in plasma bile acid profiles and the pathophysiology of T2DM.
