Solving the microheterogeneity of Bothrops asper myotoxin-II by high-resolution mass spectrometry: Insights into C-terminal region variability in Lys49-phospholipase A2 homologs

Summary: This study investigates the toxic action of Myotoxin-II, a phospholipase A(2) (PLA(2))-like protein found in Bothrops asper venom. It identifies the C-terminal region of the protein as responsible for causing rapid necrosis of muscle fibers. The study also reveals the presence of different amino acids at position 124 in variants of this toxin from the Caribbean and Pacific regions. These findings suggest that the disruptive mechanism of PLA(2)-like myotoxins toward membranes is not dependent on a fixed amino acid sequence motif, but instead on the spatial arrangement of physicochemical properties.

Distinct immunomodulatory properties of extracellular vesicles released by different strains of Acanthamoeba

Summary: This study evaluated EVs produced by Acanthamoeba from different genotypes, finding that all strains released EVs regardless of genotype, with no differences in size and/or concentration detected. EVs exhibited a predominant activity of serine proteases, with higher intensity in T4 and T1 strains. EVs from the environmental, nonpathogenic T11 strain had a more proinflammatory profile compared to strains with pathogenic traits, inducing higher levels of Nitrite, tumor necrosis factor alpha, and interleukin-6 via TLR4/TLR2. Preincubation with EVs treated with protease inhibitors or heating drastically decreased nitrite concentration production in macrophages, suggesting that the immunomodulatory effects of EVs may reflect their pathogenic potential depending on the Acanthamoeba strains and are dependent on protease integrity.
