Isolation and Profiling of Human Primary Mesenteric Arterial Endothelial Cells at the Transcriptome Level

Naseeb Kaur Malhi et al.

Summary: Understanding the transcriptome and epigenome of endothelial cells (ECs) is crucial for studying their roles in development, health, and disease. However, isolating primary ECs is challenging. Recent technologies have enabled high-throughput profiling, leading to the discovery of unknown EC cell subpopulations and developmental trajectories. This paper presents a method for isolating human primary ECs from mesenteric arteries to capture their native state, allowing for analysis of EC transcriptional and epigenetic regulation.

Long Non-coding RNA Aerrie Controls DNA Damage Repair via YBX1 to Maintain Endothelial Cell Function

Tan Phat Pham et al.

Summary: Aging is accompanied by various physiological changes, leading to cardiovascular diseases due to impaired vascular homeostasis. Increased DNA damage is a hallmark of aging, which can be repaired by the DNA signaling and repair system. In this study, it was found that long non-coding RNA Aerrie plays a role in DNA repair mechanism, affecting endothelial cell function and genomic stability.

Single-Cell Transcriptomic Atlas of Different Human Cardiac Arteries Identifies Cell Types Associated With Vascular Physiology

Zhan Hu et al.

Summary: In this study, researchers used single-cell sequencing to investigate the cellular composition of human nondiseased cardiac arteries, identifying 25 subpopulations representing 10 main arterial cell types. They found specific vascular smooth muscle cell and fibroblast subpopulations in different arteries and predicted increased intercellular communication in atherosclerosis. The results provide a cell atlas for potential identification of vascular disease-associated cell populations and development of new therapeutic strategies.

Silencing of Long Non-Coding RNA LINC00607 Prevents Tumor Proliferation of Osteosarcoma by Acting as a Sponge of miR-607 to Downregulate E2F6

Yuehuan Zheng et al.

Summary: Osteosarcoma is a malignant bone tumor commonly found in children and adolescents. In this study, it was discovered that the lncRNA LINC00607 plays a role in promoting tumor proliferation, migration, and invasion. Through interactions with miR-607, LINC00607 upregulates the expression of E2F6, leading to the progression of osteosarcoma.

RAMP2-AS1 Regulates Endothelial Homeostasis and Aging

Chih-Hung Lai et al.

Summary: This study identified a novel regulator RAMP2-AS1 essential for endothelial cell (EC) homeostasis and function, with knockdown leading to aging of vascular endothelial cells. The research demonstrated an integrative approach to quantifying EC aging based on transcriptome changes and identified other novel regulators involved in cell functional modulation.

Angiotensin II-Induced Long Non-Coding RNA Alivec Regulates Chondrogenesis in Vascular Smooth Muscle Cells

Vishnu Amaram Samara et al.

Summary: This study identified a novel AngII-induced lncRNA, Alivec, which plays a crucial role in the chondrogenic transformation of VSMCs. Alivec is co-regulated by AngII, growth factors, and inflammatory cytokines, and its knockdown attenuated the expression of chondrogenic marker genes in VSMCs. These findings suggest that AngII-regulated lncRNA Alivec is involved in the vascular dysfunction and hypertension caused by the chondrogenic transformation of VSMCs.

Emerging Role of Long Non-Coding RNAs in Diabetic Vascular Complications

Vinay Singh Tanwar et al.

Summary: Chronic metabolic disorders like obesity and diabetes are associated with accelerated vascular complications, and long non-coding RNAs play a crucial role in regulating gene expression in these diseases. Understanding the characteristics and mechanisms of lncRNAs can provide insights for the development of new therapeutic approaches to manage these disorders effectively.
