Phenols and terpenoids: natural products as inhibitors of NLRP3 inflammasome in cardiovascular diseases

Summary: Inflammatory infiltration is a factor in the development of cardiovascular diseases (CVDs), and the NLRP3 inflammasome is involved in several types of CVDs. Inhibiting the activity of the NLRP3 inflammasome can hinder the progress of CVDs. This article summarizes 20 phenols and terpenoids for the treatment of CVDs based on the inhibition of the NLRP3 inflammasome, identifying salvianolic acid B, ellagic acid, oridonin, and triptolide as potential agents for future NLRP3 inflammasome inhibitors or lead compounds.

Isoandrographolide inhibits NLRP3 inflammasome activation and attenuates silicosis in mice

Summary: This study firstly reported that (8R-12S)-isoandrographolide (ISA) from Andrographis paniculata can reduce pulmonary inflammation and fibrosis by inhibiting NLRP3, improving silicosis. ISA administration alleviated lung injury, attenuated inflammatory response, EMT, and collagen deposition in silica-induced mice. Further studies confirmed that ISA inhibited the expressions of NLRP3 inflammasome-related proteins, leading to the reduction of inflammation and EMT.
