Unmasking of CgYor1-Dependent Azole Resistance Mediated by Target of Rapamycin (TOR) and Calcineurin Signaling in Candida glabrata

Sonam Kumari et al.

Summary: In this study, the mechanisms of azole resistance mediated by CgYor1, a multidrug resistance-associated protein (MRP) subfamily ABC transporter, were analyzed. It was found that CgYor1 does not transport oligomycin but is involved in azole resistance. Under normal growing conditions, its function is masked by the major transporter CgCdr1; however, under nitrogen-depleted conditions, it displays its azole resistance function independently. It is proposed that the removal of CgYor1 affects azole susceptibility through modulation of TOR and calcineurin cascades.

Candida glabrata Antifungal Resistance and Virulence Factors, a Perfect Pathogenic Combination

Maria Guadalupe Frias-De-Leon et al.

Summary: In recent years, there has been a progressive increase in invasive fungal infections caused by Candida glabrata. Drug resistance and virulence factors are important research areas, with different mechanisms of drug resistance found for the C. glabrata complex depending on geographical regions. This complex possesses a perfect pathogenetic combination for invading immunocompromised hosts.

Deubiquitinase Ubp3 enhances the proteasomal degradation of key enzymes in sterol homeostasis

Qiuyan Lan et al.

Summary: Sterol homeostasis is tightly regulated by highly conserved molecules, with the ubiquitin-proteasome system playing a crucial role. Ubp3 enhances the proteasomal degradation of ergosterol biosynthetic enzymes, and its absence leads to the accumulation of toxic sterols, disrupting sterol homeostasis. Ubp3 may serve as a potential new antifungal target.
