Single-cell RNA profiling identifies diverse cellular responses to EWSR1/FLI1 downregulation in Ewing sarcoma cells

Roxane Khoogar et al.

Summary: In this study, the researchers investigated the transcriptional states and phenotypes of cell proliferation and dormancy by suppressing the expression of the EWSR1/FLI1 gene. They found that the dynamics of subpopulations of cells can affect the stem cell characteristics and survival mechanisms of cells, which is of significant importance for understanding the occurrence and development of Ewing sarcoma.

Combinatorial strategies to potentiate the efficacy of HDAC inhibitors in fusion-positive sarcomas

Cinzia Lanzi et al.

Summary: Fusion positive (FP) sarcomas are characterized by chromosomal rearrangements generating pathognomonic fusion transcripts and oncoproteins. HDACs play a crucial role in the pathobiology of FP sarcomas by affecting histone acetylation and chromatin remodeling, and HDAC inhibitors (HDACis) have shown potential therapeutic benefits in preclinical studies. However, their efficacy in clinical trials for solid tumors, including sarcomas, is limited. Rational drug combinations targeting specific HDAC isoenzymes may offer new opportunities for improving the therapeutic outcomes in these sarcoma histotypes.

Chromatin profiling reveals relocalization of lysine-specific demethylase 1 by an oncogenic fusion protein

Emily R. Theisen et al.

Summary: Pediatric cancers often have quiet mutational landscapes, instead characterized by single driver events like chromatin regulator mutations or oncohistone expression. The fusion protein EWS/FLI in Ewing sarcoma reshapes enhancer landscape through collaboration with LSD1, impacting widespread gene regulation throughout the genome. The dynamic rearrangement of LSD1 by EWS/FLI plays a crucial role in gene activation and establishment of super-enhancers, ultimately influencing the enhancer landscape in Ewing sarcoma.

The epigenetic basis of cellular heterogeneity

Summary: Single-cell sequencing methods have shown heterogeneity in gene expression levels among morphologically similar cells, with epigenomic information contributing to the regulation of gene expression.Recent studies suggest that variations in chromatin organization collectively define gene expression heterogeneity among otherwise similar cells.

STAG2 mutations alter CTCF-anchored loop extrusion, reduce cis-regulatory interactions and EWSR1-FLI1 activity in Ewing sarcoma

Summary: The study reveals that STAG2-LOF fine-tunes the activity of an oncogenic transcription factor by altering anchored CTCF loop extrusion and enhancer mechanisms, thereby affecting the invasive behavior of Ewing sarcoma cells.

A first-generation pediatric cancer dependency map

Neekesh Dharia et al.

Summary: A pediatric cancer dependency map was created using genome-scale CRISPR-Cas9 loss-of-function screens in 82 pediatric cancer cell lines, revealing genetic dependencies across various tumor types. Contrary to the belief that pediatric cancers have fewer mutations, it was found that genetic dependencies in pediatric cancer cell lines are similarly complex as in adult models, indicating new therapeutic targets for childhood cancers.

TRIM8 modulates the EWS/FLI oncoprotein to promote survival in Ewing sarcoma

Bo Kyung A. Seong et al.

Summary: Fusion-transcription factors (fusion-TFs) are difficult to therapeutically target, but recent research has discovered TRIM8 as a selective E3 ubiquitin ligase for degrading a driver fusion-TF in Ewing sarcoma. Knocking out TRIM8 leads to increased levels of the fusion TF, EWS/FLI, providing a potential strategy for exploiting oncogene overdose in Ewing sarcoma and other fusion-TF driven cancers.

Therapeutic Potential of EWSR1-FLI1 Inactivation by CRISPR/Cas9 in Ewing Sarcoma

Saint T. Cervera et al.

Summary: Ewing sarcoma is an aggressive bone cancer affecting children and young adults, characterized by chromosomal translocations producing chimeric oncogenic transcription factors. In this study, genetic inactivation of the EWSR1-FLI1 oncogene using CRISPR/Cas9 technology in Ewing sarcoma cells effectively blocked cell proliferation and induced a senescence phenotype. This suggests that complete inactivation of EWSR1-FLI1 at the cellular level could be a promising therapeutic approach in the future.
