The Role of the Pathogen Dose and PI3Kγ in Immunometabolic Reprogramming of Microglia for Innate Immune Memory

Trim Lajqi et al.

Summary: Microglia exhibit long-term response changes indicative of innate immune memory, with patterns of trained immunity and immune tolerance after low and high dose challenges with pathogen-associated molecular patterns, respectively. Immunometabolic reprogramming is involved in IIM in a dose-dependent manner via the glycolytic pathway, oxygen consumption and ATP production: ULP increases it, while HP reduces it.

Butyrate and Metformin Affect Energy Metabolism Independently of the Metabolic Phenotype in the Tumor Therapy Model

Felix B. Meyer et al.

Summary: The BALB/c cell transformation assay (BALB-CTA) allows for a direct comparison between untransformed and malignant cells, taking into account tumor heterogeneities. Monoclonal cell lines derived from BALB-CTA were used to study the effects of metformin and butyrate on phenotypes, revealing anti-carcinogenic effects and reduced glucose uptake in the BALB-TTM. Metabolic characterization of the cell clones showed three distinct phenotypes, with surprising opposite metabolic shifts induced by metformin or butyrate treatment across all clones.

Dynamic regulation of mitochondrial pyruvate metabolism is necessary for orthotopic pancreatic tumor growth

Nancy P. Echeverri Ruiz et al.

Summary: The study demonstrated that substitution of serine for alanine at key regulatory sites of PDHA1 generated a hypomorphic PDC, leading to reduced tumor growth; Dynamic adaptation of PDC to microenvironmental conditions is crucial for supporting pancreatic cancer growth in vivo.

Targeting the Metabolic Adaptation of Metastatic Cancer

Josep Tarrago-Celada et al.

Summary: The search for new therapeutic opportunities to target cancer metastasis is crucial for the improvement of cancer treatment. Metabolic adaptation is an important hallmark of cancer and metastasis, and targeting metabolic pathways represents a promising strategy to specifically target metastatic cells, especially in colorectal cancer metastasis. Expanding research in this field may lead to the development of new therapeutic strategies for metastatic colorectal cancer.

Trapped topoisomerase-DNA covalent complexes in the mitochondria and their role in human diseases

Benu Brata Das et al.

Summary: Topoisomerases play a crucial role in regulating DNA topology, genetic transmission, and gene expressions. Mitochondria depend on the nucleus for proteins necessary for mtDNA replication, repair, and maintenance. The mitochondrial genome faces topological stress from transcription and replication, leading to the import of nuclear encoded topoisomerases for relaxing mtDNA. Protein-DNA complexes can cause mtDNA damage and are linked to various human diseases.

Mechanistic insights into p53-regulated cytotoxicity of combined entinostat and irinotecan against colorectal cancer cells

Christian Marx et al.

Summary: Acetylation of p53, specifically at the C-terminal, controlled by HDACs, plays a crucial role in determining cellular stress responses and apoptosis in p53-positive CRC cells. The combination of irinotecan and entinostat represents clinically tractable agents for the therapy of p53-proficient CRC due to this mechanism.
