Click chemistry extracellular vesicle/peptide/chemokine nanocarriers for treating central nervous system injuries

Huitong Ruan et al.

Summary: In this research, extracellular vesicles (EVs) secreted by M2 microglia were used to improve the differentiation of neural stem cells (NSCs) at the injured site. The EVs were modified with the injured vascular targeting peptide (DA7R) and the stem cell recruiting factor (SDF-1) to recruit NSCs, induce their neuronal differentiation, and serve as nanocarriers at the injured site. The results showed that the modified EVs could target human umbilical vascular endothelial cells (HUVECs), recruit NSCs, and promote their neuronal differentiation. Furthermore, certain upregulated miRNAs in the EVs were found to induce NSCs to differentiate into neurons.

Analysis of extracellular vesicle DNA at the single-vesicle level by nano-flow cytometry

Haisheng Liu et al.

Summary: This study provides direct and conclusive experimental evidence for an in-depth understanding of how DNA is associated with extracellular vesicles (EVs). The study reveals that EVs carry naked DNA or DNA associated with non-vesicular entities. The quantity and distribution of EV-DNA vary depending on the cell type, with a high heterogeneity observed. The study also shows that external EV-DNA is mainly localized on smaller EVs, while internal EV-DNA is packaged inside larger EVs. The predominant form of EV-DNA is double-stranded DNA, and it is not associated with histone proteins.

Exosomes derived from 3D-cultured MSCs improve therapeutic effects in periodontitis and experimental colitis and restore the Th17 cell/Treg balance in inflamed periodontium

Yong Zhang et al.

Summary: Our study demonstrated that exosomes derived from mesenchymal stem cells (MSCs) cultured in a 3D system (3D-exos) have enhanced anti-inflammatory effects in periodontitis compared to those from a traditional 2D culture system (2D-exos). This improvement is mediated by the increased expression of miR-1246 in 3D-exos, which suppresses Nfat5 expression and restores the Th17 cell/Treg balance in inflamed periodontal tissues. Furthermore, local injection of 3D-exos also attenuated experimental colitis by rebalancing immune responses.

Oligodendrocytes enhance axonal energy metabolism by deacetylation of mitochondrial proteins through transcellular delivery of SIRT2

Kelly A. Chamberlain et al.

Summary: This study reveals a transcellular signaling mechanism by which oligodendrocytes deliver SIRT2 to axons to enhance energy metabolism, promoting ATP production through deacetylation of mitochondrial proteins. The transcellular delivery of SIRT2 is crucial for maintaining axonal energy enhancement, providing a potential target for boosting bioenergetic metabolism in neurological disorders. Small extracellular vesicles released by infused mesenchymal stromal cells target M2 macrophages and promote TGF-β upregulation, microvascular stabilization and functional recovery in a rodent model of severe spinal cord injury

Masahito Nakazaki et al.

Summary: Intravenous infusion of bone marrow-derived mesenchymal stem/stromal cells (MSCs) leads to the release of small extracellular vesicles (sEVs) that can target the injury site in spinal cord injury (SCI) rats and interact with M2 macrophages. Fractionated dosing of MSC-sEVs over 3 days is required to achieve therapeutic effects, mimicking the results of single MSC infusion.
