Clinicopathological features of syncytial variant nodular sclerosis Hodgkin lymphoma

Summary: SV-NSHL is a rare subtype of Hodgkin lymphoma, typically presenting in younger patients with mediastinal masses and high relapse rates. Immunohistochemical analysis revealed positivity for several markers, with EBER detected in some cases. Factors associated with poorer overall survival included age, B-type symptoms, elevated LDH levels, and disease relapse, while EBER-positive tumors were linked to better survival outcomes. Compared to non-SV-NSHL patients, those with SV-NSHL had worse overall survival, indicating a poorer prognosis with traditional therapy.

Summary: High PD-L1 protein expression and CNAs of 9p24.1 in HRSC are associated with favorable response to anti-PD-1 therapy in cHL. However, in early stage unfavorable cHL, 9p24.1 CNAs and PD-L1/MHC-I/II expressions did not impact early response to anti-PD-1 treatment. Further studies are needed to evaluate an expanded panel of potential biomarkers for optimal stratification of first-line anti-PD-1 treatment in cHL.

The Hodgkin Lymphoma Immune Microenvironment: Turning Bad News into Good

Summary: This article reviews the important immune populations in the cHL tumor microenvironment, focusing on their integrative functional signatures. The cHL microenvironment plays a significant role in tumor cell survival. HRS cells promote their survival through intrinsic and extrinsic alterations, leading to immunosuppression. cHL is proving to be an ideal model for studying immune checkpoint blockade, with druggable key signaling pathways identified. Additionally, numerous biomarkers for predicting treatment response await further validation.

The clinicopathological and molecular features of sinusoidal large B-cell lymphoma

Summary: This study presented 17 cases of sinusoidal large B-cell lymphoma (SLBCL) with distinct pathological features. The patients had a median age of 57 years, with approximately equal numbers of male and female patients. The majority of patients exhibited aggressive disease and poor outcomes.

Summary: This study reported 17 cases of H/DC neoplasms in Japanese patients, with two cases of HS and one IDC sarcoma exhibiting t(14;18). Additionally, 30% of FDC sarcoma cases showed neoplastic PD-L1 expression. This is the first report of t(14;18) and neoplastic PD-L1 expression on H/DC neoplasms among Japanese patients.

Single-cell profiling reveals the importance of CXCL13/CXCR5 axis biology in lymphocyte-rich classic Hodgkin lymphoma

Summary: LR-CHL has a unique CD4* helper T cell subset characterized by high expression of CXCL13 and PD-1. PD-1*CXCL13* T cells are significantly enriched in LR-CHL compared to other CHL subtypes, and their abundance in the TME is significantly associated with shorter progression-free survival. These findings suggest the importance of the CXCL13/CXCR5 axis and PD-1*CXCL13* T cells as a treatment target in LR-CHL.

Summary: Primary adrenal diffuse large B-cell lymphoma (PA-DLBCL) is a rare disease with unique clinicopathologic features and biological behavior. Patients with PA-DLBCL can be classified into different subgroups, and significant differences in prognosis were observed among these subgroups. Factors such as immune senescence, iatrogenic immunodeficiency, and immune evasion were found to contribute to the development of PA-DLBCL.
