Mining of a novel esterase (est3S) gene from a cow rumen metagenomic library with organosphosphorus insecticides degrading capability: Catalytic insights by site directed mutations, docking, and molecular dynamic simulations

Hee Yul Lee et al.

Summary: A novel esterase gene (est3S) was cloned from uncultured microorganisms in cow rumen, with the Est3S protein showing enhanced activity with K+, Na+, Mg2+, Ca2+, and the ability to degrade chlorpyrifos and methyl parathion. A mutated Est3S (Ser132-Ala132) showed no activity towards chlorpyrifos and esters, indicating the importance of Ser132 as a critical amino acid. The Est3S protein did not fall into any established classes of esterase/lipase proteins, and it exhibited stability with ester compounds during molecular dynamics simulations.
