Summary: The study found that high expression levels of cGAS and IFI16 in PBMC from SLE patients were strongly correlated with disease activity. Both cGAS and IFI16 mediated signaling pathway contributed to the robust production of IFN-beta. Expression of cGAS and IFI16 combined with IFN-beta in PBMC might serve as potential biomarkers for early diagnosis and monitoring disease activity in SLE.

Type I Interferons in Autoimmunity

Summary: Dysregulated IFN-1 responses are important in the development of various autoimmune diseases. Enhanced IFN-1 signaling is seen in patients with lupus, systemic sclerosis, Sjogren's syndrome, and dermatomyositis. Excess IFN-1 is associated with disease severity and autoantibodies, and may be used to predict response to new therapies targeting IFN-1 pathways. This review provides an overview of IFN-1 signaling pathway and immune functions in both health and disease, as well as current therapeutic strategies targeting the IFN-1 system.
