Massive tramadol ingestion resulting in fatal brain injury - a pharmacokinetic study with discussion on the involved mechanisms of toxicity

Alicja Puszkiel et al.

Summary: This study investigated the mechanisms of toxicity in a case of massive tramadol ingestion. A female patient who ingested 9000 mg sustained-release tramadol developed severe brain injury. The study found that the plasma concentrations of tramadol and its metabolites were extremely high, and the elimination half-lives were prolonged, possibly involving cerebral vasospasm.

In vivo and in vitro α-amanitin metabolism studies using molecular networking

Brendan Le Dare et al.

Summary: Amanitin poisonings caused by the genus Amanita are some of the most life-threatening mushroom poisonings, mainly characterized by hepatotoxicity due to the powerful toxins contained in these mushrooms. Through molecular networking, it has been demonstrated that alpha-amanitin does not undergo metabolism in humans, providing solid evidence that amanitin metabolites are not involved in its toxicity pathways. These findings can help to settle the debate on amanitin metabolism and toxicity.
