In Situ Super-Resolution Imaging of Organoids and Extracellular Matrix Interactions via Phototransfer by Allyl Sulfide Exchange-Expansion Microscopy (PhASE-ExM)

Applications of Super Resolution Expansion Microscopy in Yeast

A Universal Labeling Strategy for Nucleic Acids in Expansion Microscopy

Summary: Expansion microscopy (ExM) allows nanoscale imaging of RNA on a conventional fluorescence microscope. A series of multivalent reagents were examined for labeling DNA probes, showing compatibility with RNA FISH techniques and retention of targeted transcripts, as well as validation for multicolor staining. Oligonucleotide-conjugated antibodies demonstrated excellent performance in ExM, achieving resolutions of around 50 nm. Our results suggest that multivalent molecules enable rapid functionalization of DNA oligonucleotides for ExM.

Multiplexed single-cell profiling of chromatin states a genomic loci by expansion microscopy

Summary: A new method called SCEPTRE was developed to visualize and quantify multiple histone modifications in nonrepetitive genomic regions of single cells using Expansion Microscopy (ExM). SCEPTRE enables the differentiation of multiple histone modifications at single gene loci, quantification of modification levels at different genes in individual cells, and evaluation of the relationship between histone modifications and RNA polymerase II loading. This technique reveals extensive variability in epigenetic states between individual gene loci, which is often concealed in current population-averaged measurements.
