Adsorption Kinetics and Self-Assembled Structures of Aspergillus oryzae Hydrophobin RolA on Hydrophobic and Charged Solid Surfaces

Summary: The adsorption kinetics of hydrophobin RolA onto solid surfaces and the self-assembled structures formed by RolA molecules were studied. The results showed that the amount of adsorbed RolA, the chemical properties of the surfaces, and the zeta potential of RolA all affected the self-assembled structures formed on the surfaces.

Summary: Aspergillus fumigatus, a major pathogenic fungus, is known to cause life-threatening infections. This study investigates the effect of isoeugenol on the hydrophobicity and biofilm formation of A. fumigatus. The results show that isoeugenol reduces hydrophobicity and biofilm formation, as well as downregulates the expression of genes related to these processes. Further studies are needed to explore the potential of isoeugenol as a treatment for A. fumigatus infections.

However, challenges in genetic manipulation and varying expression levels of heterologous proteins have hindered its further development and application. Recent advances in CRISPR genome editing and high-throughput screening platforms have facilitated modifications and applications of Aspergillus. Multi-omics analysis and multiplexed genetic engineering have also promoted effective knowledge mining.

Orientation and Conformation of Hydrophobin at the Oil-Water Interface: Insights from Molecular Dynamics Simulations

Summary: Hydrophobins are widely used in the food, pharmaceutical, and chemical industries as potential protein emulsifiers. This study employed molecular dynamics simulations to investigate the adsorption orientation and conformation change of hydrophobin HFBI at the oil-water interface. The results showed that HFBI adsorbed at the interface with a patch-up orientation towards the oil phase, maintaining its secondary structure intact. The driving force for adsorption was primarily determined by van der Waals interactions, while the adsorption orientation and conformation were regulated by hydrophobic patches and key residues.

Fungal Hydrophobin RolA Enhanced PETase Hydrolysis of Polyethylene Terephthalate

Summary: Polyethylene terephthalate (PET) is a well-known polyester used as packaging material, and PETase has emerged as a potential biocatalyst for degrading PET. The presence of hydrophobin RolA significantly increased the rate of PETase hydrolysis, while enhancing the hydrolysis of PET bottles. This study suggests that RolA's wetting effect on the PET surface may convert PET to become hydrophilic, making it easier for PETase to contact and attack the surface.

Class I hydrophobin fusion with cellulose binding domain for its soluble expression and facile purification

Summary: Hydrophobins are highly surface-active proteins that can reverse surface hydrophobicity through self-assembly at interfaces, with potential applications in surface functional modifications. By fusing cellulose-binding domain with hydrophobins, soluble expression in E. coli can be enhanced for rapid isolation and purification. This strategy also retains the interfacial activity of hydrophobins and accelerates PETase hydrolysis against PET fiber.

Optimization and scale-up of α-amylase production by Aspergillus oryzae using solid-state fermentation of edible oil cakes

Summary: The study successfully produced alpha-amylase using Aspergillus oryzae and low-cost substrates obtained from edible oil cake through solid-state fermentation. Optimization of process parameters led to the identification of groundnut oil cake as the most suitable substrate for maximizing enzyme production. The upregulated genes during infection of chickpea lines with different resistance levels included carbohydrate-active enzymes, secondary metabolite biosynthesis clusters, transcription factors, and candidate secreted effectors. These findings provide insights into the molecular interactions between S. sclerotiorum and hosts of varying susceptibility levels.

Engineered Hydrophobin as a Crystallization Inhibitor for Flufenamic Acid

Summary: This study evaluated an engineered, recombinant hydrophobin as a potential crystallization inhibitor for maintaining drug supersaturation in an amorphous drug delivery system. Results showed that HFB1 outperformed all tested polymers, potentially offering a more effective and lower concentration option for crystallization inhibition in amorphous drug delivery systems. Dynamic light scattering and circular dichroism results suggested a mechanism in which HFB1 functioned differently depending on the state of the drug.
