Evolution of biofilm-adapted gene expression profiles in lasR-deficient clinical Pseudomonas aeruginosa isolates

Alexander Jeske et al.

Summary: This study analyzed the transcriptional profiles and genomic sequence variations of clinical Pseudomonas aeruginosa isolates to uncover convergent changes in gene expression patterns. A defective IasR gene was found to be the cause of the majority of observed expression patterns, and lasR-deficient strains exhibited a transcriptional response consistent with biofilm growth without requiring an environmental trigger. These findings provide insights into the long-term evolutionary pathways and success of lasR mutants in clinical P. aeruginosa.

Pseudomonas aeruginosa Strain-sharing in Early Infection Among Children With Cystic Fibrosis

Patrick J. Stapleton et al.

Summary: In early CF infections, there is evidence of P. aeruginosa strain-sharing, with some infections likely being transmitted between patients. Mixed-strain infections are associated with strain-sharing, but strain-sharing does not impact antibiotic eradication treatment failure rates.

Omics-based tracking of Pseudomonas aeruginosa persistence in eradicated cystic fibrosis patients

Jennifer A. Bartell et al.

Summary: The study reveals that some CF patients may continue to be infected with Pseudomonas aeruginosa even after antibiotic therapy, suggesting the need for tailored antibiotic treatment. It also finds that most patients' infections are associated with surgery, and persistent infection is due to the re-emergence of the same strain rather than re-infection from the environment.

The Rhl quorum sensing system is at the top of the regulatory hierarchy under phosphate limiting conditions in Pseudomonas aeruginosa PAO1

Summary: This study reveals that under phosphate limitation, even in the PAO1 type strain, the absence of LasR has no effect on the production of virulence factors and gene expression profile, whereas in a condition of phosphate repletion, the LasR hierarchy is maintained.

Pseudomonas aeruginosa OprF plays a role in resistance to macrophage clearance during acute infection

Malika Moussouni et al.

Summary: The study demonstrates that a P. aeruginosa oprF mutant is attenuated in zebrafish embryos with functional macrophages, but not due to increased macrophage recruitment or enhanced phagocytosis. Additionally, inhibition of acidification increases embryo sensitivity to both wild-type and oprF mutant, partially reversing the attenuation caused by oprF mutation.

Pseudomonas aeruginosa aggregates in cystic fibrosis sputum produce exopolysaccharides that likely impede current therapies

Laura K. Jennings et al.

Summary: In cystic fibrosis airways, Pseudomonas aeruginosa forms biofilms that are thought to contribute to chronic infection. Exopolysaccharides Pel and Psi are expressed in CF airways and play a role in the aggregation mechanism, potentially impacting therapies by increasing antimicrobial tolerance and protecting extracellular DNA from digestion.

Inhibitor Mimetic Mutations in the Pseudomonas aeruginosa PqsE Enzyme Reveal a Protein-Protein Interaction with the Quorum-Sensing Receptor RhlR That Is Vital for Virulence Factor Production

Isabelle R. Taylor et al.

Summary: The study identified a new route for drug discovery targeting PqsE in Pseudomonas aeruginosa, involving screening molecules that bind the virulence-controlling protein PqsE and characterizing hit compounds for inhibition of PqsE enzymatic activity. The results suggest that interactions between PqsE and the quorum-sensing receptor RhlR are responsible for promoting virulence factor production in P. aeruginosa.

Outer membrane permeability: Antimicrobials and diverse nutrients bypass porins in Pseudomonas aeruginosa

Johanna Ude et al.

Summary: This study found that all porins in Pseudomonas aeruginosa were not essential for bacterial growth in rich medium and consumption of diverse hydrophilic nutrients. However, nutrients with multiple carboxylate groups permeated poorly in the absence of porins. Porins provided efficient pathways for nutrient uptake while effectively excluding most antibiotics except carbapenems, with outer-membrane lipid bilayer serving as a major pathway for nutrient and drug entry into the bacterial cell.

Summary: Therapy for Pseudomonas aeruginosa infections is challenging due to its high antibiotic resistance. The study reveals that hyperexpression of an efflux pump MexAB-OprM caused by a R70Q substitution in MexR contributes to increased resistance to aztreonam in clinical isolates. This finding sheds light on the mechanism of antibiotic resistance development in P. aeruginosa.

Summary: This study aimed to investigate the prevalence of mutations upstream of the oprD coding region and its promoters in imipenem-resistant and sensitive Pseudomonas aeruginosa isolated from educational hospitals in Yazd City, Iran. Mutations were observed in the upstream regions of the oprD gene in the majority of resistant isolates, suggesting a potential mechanism for imipenem resistance. The findings emphasize the importance of controlling the indiscriminate use of antibiotics to prevent the development of resistance.
