Differential lipidomics of HK-2 cells and exosomes under high glucose stimulation

Weidong Wang et al.

Summary: Abnormal lipid composition and differential expression of renal tubular cells and their exosomes under high glucose stimulation play an important role in the development of diabetic kidney disease. This study identified specific lipid species and revealed their changes, providing new targets for exploring the mechanisms of DKD.

Pathophysiology of Lipid Droplets in Neuroglia

Tina Smolic et al.

Summary: Recent evidence has highlighted the functional importance of lipid droplets (LDs) in the central nervous system (CNS). LDs play a crucial role in energy production, membrane synthesis, and cellular protection by storing and releasing fatty acids, cholesterol, and ceramides. Accumulation of LDs in neuroglia provides support for neuronal networks.

Extracellular Vesicles in Redox Signaling and Metabolic Regulation in Chronic Kidney Disease

Omar Emiliano Aparicio-Trejo et al.

Summary: Chronic kidney disease (CKD) is a rapidly increasing global health problem, with metabolic reprogramming and changes in redox signaling playing crucial roles in its development. Extracellular vesicles (EVs) play a significant role in the spread of renal injury in CKD and are involved in redox signaling and metabolic reprogramming. Understanding the role of EVs and their cargos in CKD can help prevent its progression.

Resveratrol Butyrate Ester Protects Adenine-Treated Rats against Hypertension and Kidney Disease by Regulating the Gut-Kidney Axis

Chien-Ning Hsu et al.

Summary: This study investigated the effectiveness of resveratrol butyrate ester (RBE) in protecting against hypertension and kidney damage in a rat model of chronic kidney disease (CKD). The results showed that RBE had a protective effect and could modulate gut microbiota composition and expression of short chain fatty acid receptors.

Flazin as a Lipid Droplet Regulator against Lipid Disorders

Xunzhi Wu et al.

Summary: In this study, the effect of flazin on lipid regulation was investigated at both the cell level and organelle level. The results showed that flazin significantly decreased cellular triglyceride and neutral lipid content, reduced lipid droplet size, and improved lipid droplet morphology. These findings suggest that flazin may be a potential therapeutic approach for treating lipid metabolism disturbances and contribute to the development of other nutraceuticals for related metabolic diseases.

Comprehensive metabolomic study of the response of HK-2 cells to hyperglycemic hypoxic diabetic-like milieu

Alberto Valdes et al.

Summary: The study revealed that the combined exposure of high glucose and hypoxia significantly altered multiple metabolic pathways in HK-2 cells, including carbohydrate, pentose phosphate, arginine, and proline metabolism. Other affected metabolites were found in purine and lipid metabolism, as well as in protecting against osmotic stress and preventing the activation of the beta-oxidation pathway. Overall, the effects observed are consistent with previous in vivo studies.

Lipophagy deficiency exacerbates ectopic lipid accumulation and tubular cells injury in diabetic nephropathy

Yachun Han et al.

Summary: The deficiency of autophagy-mediated lipophagy plays a critical role in the ectopic lipid deposition and renal injury related to lipids in diabetic nephropathy (DN). Decreased lipophagy, increased ectopic lipid deposition and lipotoxicity were observed in tubular cells of DN patients and db/db mice, and these effects could be reversed by the autophagy-promoting adiponectin receptor activator AdipoRon.

Summary: By using chemiluminescence detection-high performance liquid chromatography instrument and liquid chromatography-tandem mass spectrometry, researchers are able to selectively detect and quantify lipid peroxidation products, as well as discriminate the position and stereoisomerization of hydroperoxide groups in lipid hydroperoxides. These methods have confirmed the reaction mechanisms of lipid peroxidation in food and in the body.

Sugar or Fat? Renal Tubular Metabolism Reviewed in Health and Disease

Leslie S. Gewin

Summary: The kidney relies on specialized epithelial cells in the renal tubules for reabsorption, with the proximal tubules using fatty acid oxidation as the preferred metabolic pathway. After kidney injury, there are alterations in metabolism leading to decreased fatty acid oxidation and increased lactic acid generation.

Adipose triglyceride lipase protects renal cell endocytosis in a Drosophila dietary model of chronic kidney disease

Aleksandra Lubojemska et al.

Summary: This study demonstrates that high-fat diet-induced renal lipid droplet accumulation can impair key functions of renal cells. The findings suggest that fatty acid flux through the lipid droplet triglyceride compartment can protect renal cells from harmful effects of excess lipid accumulation. Boosting expression of the lipid droplet resident enzyme adipose triglyceride lipase is sufficient to rescue defects in renal endocytosis, with a requirement for the mitochondrial regulator PGC1 alpha.
